摘 要:目的:研究健脾法在抗肿瘤作用中与调节非特异性免疫相关的机制。方法:采用人胃癌细胞株MKN-45裸小鼠皮下移植瘤模型,随机分为全方组、健脾组、非健脾组、5-FU组和生理盐水组以及非造模组。观察抑瘤率,乳酸脱氢酶(LDH)释放法检测自然杀伤(NK)细胞杀伤活性、ELISA方法检测血清γ干扰素(IFN-γ)含量、Western Blot检测脾细胞NKG2D受体表达。结果:全方组、健脾组、非健脾组和5-FU组平均瘤重均低于生理盐水组(P<0.01)。全方组、健脾组、非健脾组NK细胞杀伤活性、血清IFN-γ含量、NKG2D受体表达量分别高于生理盐水组(P<0.01),5-FU组则无变化;健脾组与全方组相比,上述指标均无统计学差异;非健脾组NK细胞杀伤活性及血清IFN-γ含量均低于全方组(P<0.01);非健脾组NKG2D受体表达量与全方组无统计学意义。结论:以健脾为基础的复方对MKN-45皮下移植瘤小鼠具有抑瘤和增强宿主非特异性免疫的作用,且健脾组分在其中起主导作用。Objectives: To investigate the method of invigorating spleen in tumor growth-suppression and modulating non-specific immunity. Methods: BALB/c mice were planted MKN-45 cancer cell blunk at armpit to establish experimental model, after that were randomizedly divided into compound recipe group, invigorating spleen group, non-invigorating spleen group, 5-FU group, NS group and non-tumor model group. Observation of diverse effects on model mice treated by different means, tumor suppression were investigated by summed the average tumor blunk weight and calculate inhibition rate; non-specific immune function change were investigated through detecting NK cell killing ability, IFN-7 capacity in sera and NKG2D expression on spleen cells. Results: The average tumor weight of compound recipe group, invigorating spleen group, non-invigorating spleen group and 5-FU group were lower than NS group (P〈0.01), showing significant difference. The NK cell killing ability, the IFN-3, capacity in sera and the NKG2D expression on spleen ceils in compound recipe group, invigorating spleen group and non- invigorating spleen group were all higher than that in NS group (P〈0.01), showing significant difference; the NK cell killing ability, the IFN-y capacity in sera and the NKG2D expression on spleen cells in 5-FU group compared with that in NS group, there was no significant difference; invigorating spleen group compared with compound recipe group, there was no significant difference; the NK cell killing ability and the IFN-y capacity in sera in non-invigorating spleen group were all lower than that in compound recipe group (P〈0.01), showing significant difference; the NKG2D expression on spleen cells in non-invigorating spleen group compared with that in compound recipe group, showing no significant difference. Conclusion: The compound recipe based on invigorating spleen has the function of tumor growth-suppression and enhance non-specific immunity in MKN-45 tumor bearing mice, and invigorating spleen components
