心复康口服液对心肌梗死后心力衰竭大鼠血浆代谢组学影响的研究

摘　　要：目的研究心复康口服液对心肌梗死后心衰大鼠血浆代谢组学的影响。方法采用结扎SD大鼠前降支复制心肌梗死后心衰模型,以心复康口服液于造模后72 h灌胃给药,以只穿线而不结扎前降支为假手术对照组。给药4周后以1H-NMR技术检测大鼠血浆小分子及大分子代谢物,并采用OPLS-DA对代谢图谱进行主成分分析及模式判别,以观察心复康口服液对心衰大鼠代谢物变化的影响。结果模型组与假手术组、心复康组与模型组代谢轮廓均能明显区分。与假手术组比较,模型组丙氨酸、乳酸、3-羟基丁酸、亮氨酸、缬氨酸、蛋氨酸、肌酸、牛磺酸、氧化三甲胺、磷脂酰胆碱含量升高;葡萄糖、N-乙酰糖蛋白、不饱和脂肪酸、脂肪酸、高密度脂蛋白、极低密度脂蛋白/低密度脂蛋白含量下降。与模型组比较,心复康组丙氨酸、3-羟基丁酸、肌酸、亮氨酸、缬氨酸、牛磺酸、磷脂酰胆碱含量下降;N-乙酰糖蛋白、葡萄糖、乳酸、不饱和脂肪酸、极低密度脂蛋白/低密度脂蛋白含量升高;N-氧化三甲胺、高密度脂蛋白、脂肪酸含量无明显变化。结论心复康口服液能有效调节心衰大鼠糖代谢、脂代谢、氨基酸代谢。Objective To study the effect of Xinfukangoral liquid （XFK） on the plasma metabolic profiling in heart failure rats after myocardial infarction. Methods The model of heart failure after myocardial infarction was established with SD rat by ligaturing the anterior descending branch, and intragastric administration with XKF was applied after 72h of modeling, while the sham operation group was set up only with threading. After 4 weeks of application, the micromolecular and macromolecular metabolites of rat plasma were detected with 1H-NMR, moreover principal component analysis and pattern discrimination were conducted for metabolic profile with OPLS-DA, so as to observe the effects of XFK on the change of metabolites in rats with heart failure. Results The metabolic profiling could be obviously distinguished in model group with sham operation group, XFK group with model group. Compared with the sham operation group, the contents of alanine, lactic acid, 3-hydroxybutyric acid, leucine, valine, methionine, creatine, taurine, trimethylamine oxide, and phosphatidylcholine in the model group were increased; while of glucose, N-acetylglycoproteins, unsaturated fatty acid, fatty acid, high density lipoprotein, very low density lipoprotein and/or low density lipoprotein decreased. Compared with the model group, the contents of alanine, 3-hydroxybutyric acid, creatine, leucine, valine, taurine and phosphatidylcholine in the XFK group were lowered; of N-acetylglycoproteins, glucose, lactic acid, unsaturated fatty acid, very low density lipoprotein and/or low density lipoprotein increased; while N-trimethylamine oxide, high density lipoprotein, fatty acid had no significant difference. Conclusion XFK could effectively regulate the disorders of glycometabolism, lipid metabolism, and amino acid metabolism in rats with heart failure.
