局部应用甲状腺素促进周围神经再生的实验研究

摘  要:【目的】观察甲状腺素对大鼠损伤的外周神经元再生的影响。【方法]60只成年健康SD大鼠,随机分成两组,每组30只。对照组和实验动物复制大鼠坐骨神经完全切断模型后,对照组离断桥接处局部给予生理盐水,实验组给予三碘甲状腺原氨酸(T3)。通过观察术后3、9、16周缩足反射刺激强度、运动神经元传导速度和S-100染色阳性值来评价外周神经再生情况。【结果】术后9、16周:①与对照组相比,实验组大鼠引起缩足反射所需的刺激强度明显更小,且差异有显著性(P〈0.05);②与对照组相比,实验组大鼠运动神经元传导速度明显更快,且差异有显著性(P〈0.05);③与对照组相比,实验组大鼠桥接处神经s-100染色阳性值明显增加,且差异有显著性(P〈0.05)。【结论】在损伤的坐骨神经局部给予T3,可以加快坐骨神经的感觉功能和运动神经元传导速度的恢复,促进神经膜细胞的再生。[Objective]To observe the effect of thyroid hormone on the regeneration of injured peripheral nerve in rats. [Methods]Sixty healthy adult SD rats were randomly divided into two groups with 30 rats in each group. Rat models were copied by complete transaction of sciatic nerve. The control group was given normal saline at the site of bridge junction, while the experimental group was given thyroid hormone triiod9thyronine(T3 ). The regeneration of peripheral nerve was estimated by observing the withdrawal reflex stimulus intensity, conduction velocity of motor neurons and the positive value of S-100 staining on 3, 9 and 16 weeks after operation. [Results]Compared with con- trol group, the withdrawal reflex stimulus intensity in experimental group on 9 and 16 week after operation was obvi- ously smaller, and there was significantly difference( P d0.05). Compared with control group, the conduction veloc- ity of motor neurons in experimental group was obviously faster, and there was significant difference( P d0. 05). Compared with control group, the positive value of S-100 staining at bridge junction in experimental group was obvi- ously increased, and there was significant difference( P d0. 05). [Conclusion]Thyroid hormone locally given in in- jured sciatic nerve can accelerate the recovery of sensory function of sciatic nerve and conduction velocity of motor neu- rons, and promote the regeneration of Schwann cells.
