摘 要:【目的】探讨不同浓度的卵蛋白(0VA)泵雾化激发方式对大鼠哮喘模型的影响。【方法】将32只SD大鼠按照0VA激发浓度的不同分为4组:用PARIBOY高频雾化器激发浓度分别为0%、1%、2%、4%(O%浓度为对照组,激发以生理盐水代替)。各0VA先按常规建立哮喘模型。各组实验大鼠于末次激发24h后(即d。,)麻醉,采用BUXCO测定气道阻力。取右肺组织做HE、Masson和AB-PAS染色检测,分析气道炎症和气道重塑情况;做左肺支气管肺泡灌洗(BAL)后,行其灌洗液(BALF)细胞学分析。【结果】①一般观察:OVA各组大鼠均出现呼吸急促、点头状呼吸、腹肌痉挛等表现,而对照组大鼠激发前后一般观察无明显变化,呼吸平稳,运动敏捷。②与对照组相比较,0VA各组支气管周围炎性细胞浸润评分、支气管管壁周围E0s、气道平滑肌面积、杯状细胞占上皮细胞百分比、胶原沉积面积显著增高(P〈0.01);OVA各组之间无显著差异(P〉0.05)。③相对于对照组,0VA各组BALF细胞总数、各细胞分类中嗜酸性粒细胞(EOS),淋巴细胞显著增高(P〈0.01);但0VA各组之间无显著差异(P〉0.05)。④OVA各组大鼠RflW值均大于对照组(P〈0.01);而0VA各组之间无显著差异(P〉0.05)。【结论】①不同浓度泵雾化激发均能够成功复制哮喘大鼠模型。②在一定范围下,卵蛋白泵雾化激发浓度对模型制作无明显影响。[Objective] To explore the effect of atomized challenge methods with different concentration of ovalbumin (OVA) pump on rat model with asthma. [Methods]Thirty-two male SD rats were divided into 4 groups. According to the concentration of OVA challenge, pump atomization group was divided into 3 groups with 8 rats in each, and the challenge concentration was 1%, 2% and 4%, respectively. After sensitization, each pump group inhaled 1%, 2% and 4% OVA for the challenge by using PARI BOY high-frequency atomizer. The control group( n =8) was challenged by normal saline. The rats in each group were anesthetized at 24h after the last challenge(57th day). BUXCO was used to measure airway pressure. Right lung tissue was removed for HE, Masson and AB-PAS staining. Airway inflammation and remodeling were analyzed. Bronchoalveolar lavage of left lung was performed for BALF cytology analysis. [Results] General observation of the rats in each group showed tachypnea, nodded-like breath and abdominal muscle spasro. General observation of the rats in control group showed stable breath and nimble movement without obvious change before and after challenge. Compared with the control group, the score of inflammatory cell infiltration around bronchus, EOS around bronchial wall, airway smooth muscle area, the percentage of goblet cells in epithelial cells and collagen deposition area in each pump group significantly increased( P 〈0.01), but there was no significant difference among pump groups( P 〉0. 05). Compared with the control group, the number of BALF cells, EOS in all various cells and lymphocytes in each pump group significantly increased( P 〈0. 01), but there was no significant difference among pump groups( P 〉0.05). Raw value in rats of each pump group was higher than that in control group( P 〈0. 01), but there was no significant difference among pump groups( P 〉0. 05).
