摘  要:目的探讨TAK1对TGF-β1诱导小鼠巨噬细胞凋亡的影响。方法电穿孔法将TAK1等目的基因质粒瞬时转染小鼠巨噬细胞RAW264.7,pEGFP-CⅡ观察转染效率,采用Real time-PCR及Western blot检测RAW264.7细胞转染后TAK1基因和蛋白的表达水平,流式细胞术检测过表达TAK1后对RAW264.7细胞凋亡的影响。结果 TAK1转染组、TAK1+TAB1共转染组细胞凋亡率明显低于空质粒对照组。结论巨噬细胞中过表达TAK1能抑制TGF-β1刺激所诱导的凋亡,且共转染TAB1+TAK1组抑制作用最明显。Transforming growth factor β-activated kinase 1(TAK1),a member of the MAPKKK family,is a key mediator of pro-inflammatory and stress signal,which plays important roles in mediating inflammation,B cell activation,and epithelial cell survival.Here,we investigated the essential function of TAK1 in the process of apoptosis of mouse macrophage cells.Plasmids of TAK1 were transfected into RAW264.7 cells,then the expression levels of TAK1 mRNA and TAK1 protein were detected by real-time PCR and Western blot respectively,and the apoptosis of RAW264.7 cells transfected with the plasmids of TAK1 or TAK1+TAB1 was evaluated by FACS.Our data showed that the level of TAK1 was significantly increased in the groups which were transfected the plasmids of TAK1 or TAK1+TAB1,and the rate of apoptosis in these groups was significantly lower,as compared with control group.These results suggest that overexpression of TAK1 can protect macrophage cells from TGF-β1-induced apoptosis.Furthermore,TAB1 interaction with TAK1 may be crucial for the activation of TAK1 signaling in TGF-β1-induced macrophage apoptosis.
