翻白草油对呼吸道合胞病毒感染宿主HeLa细胞凋亡的影响

摘　　要：目的探讨翻白草油抗呼吸道合胞病毒(RSV)的作用。方法 RSV感染宿主HeLa细胞后,进行翻白草油干预,采用流式细胞术检测宿主HeLa细胞凋亡百分比;Western印迹和免疫荧光检测caspase-3蛋白的表达。结果翻白草油(除药物预处理作用)组与RSV病毒组比较,宿主HeLa细胞凋亡百分比及caspase-3蛋白的表达明显降低(P<0.01)。结论翻白草油在三种作用方式下(直接灭活阶段、病毒复制阶段、病毒吸附阶段)有抗RSV作用,可能与降低宿主HeLa细胞凋亡及caspase-3蛋白的表达有关。Objective To make sure the anti-RSV effect of Potentilla discolor oil.Methods The expression of caspase 3 was detected by Western blot and the apotosis rate was determined by FACS in Hela cells infected with RSV,the protein was extracted to detect after Potentilla discolor oil intervention.Results Compared with the virus control group,the expression of caspase-3 protein and the apotosis rate decreased obviously in Potentilla discolor oil group（P0.01）.Conclusions Potentilla discolor oil has anti-RSV effect by direct inactivation,inhibition of virus replication and virus absorption.The anti-RSV effect of Potentilla discolor oil is related to decreasing the expression of caspase-3 protein and the apoptosis rate of Hela cells infected with RSV possibility.
