耻骨联合软骨细胞与藻酸盐凝胶复合物体内培养研究

摘　　要：目的 观察耻骨联合软骨细胞与藻酸盐凝胶支架复合物体内异位培养的情况.方法 从SD大鼠耻骨联合组织分离细胞体外培养后,按5×10^7/ml细胞密度与1.2%（w/v）低黏度海藻酸钠溶液混合,制备细胞凝胶复合物;再将细胞凝胶复合物植入裸鼠皮下,6周和12周后取材;通过HE、阿尔新蓝和Ⅱ型胶原免疫组化染色,观察体内异位形成软骨的情况.结果 耻骨联合软骨细胞分散悬浮在凝胶支架中.6周和12周的裸鼠皮下标本见复合物中有新生的岛样组织出现,12周标本岛样组织明显增多,阿尔新蓝和Ⅱ型胶原免疫组化染色显示岛样组织基质阳性显色.结论 耻骨联合软骨细胞与藻酸盐凝胶支架复合后,在体外仍然保持细胞活性,并且在裸鼠体内具有一定的异位软骨形成的能力.Objective To observe the heterotopic cultivation of chondrocyte from pubic symphysis and alginate gelatin compound in vivo. Methods Cells isolated from pubic symphysis cartilage were cultured in vitro, the 5 ×10^7/ml cultured cells were mixed with 1.2% （w/v） alginate sodium solution to form the cell-gelat- in compound and then the compound was implanted subcutaneously into the nude mouse. The samples were harvested at 6 weeks and 12 weeks, the heterotopic chondrification was observed by HE, aleian blue and typeⅡ collagen immunohistochemical staining. Results Cells isolated from pubic symphysis cartilage were dispersed and suspended in gelatin scaffold and island-like tissue was observed in the compound of nude mouse at 6 weeks and 12 weeks, and more at 12 weeks. The island-like tissue was positive in alcian blue and collagen type Ⅱ immunohistochemical staining. Conclusion The compound of chondroeyte from pubic symphysis and alginate gelatin scaffold shows cell activity in vivo and are capable of heterotopic chondrification.
