摘  要:为建立基因C型鸭甲型肝炎病毒(DHAV-C)在鸭胚原代肝细胞(DELC)上的感染模型,利用17日龄鸭胚制备了DELC,接种DHAV-C,观察接毒后DELC的病变情况,并采用荧光定量PCR方法对病毒含量进行测定,绘制病毒增殖曲线。结果显示,DELC在接毒后第36小时开始出现细胞病变,第60小时细胞病变最为明显,表现为细胞间隙增大、拉网成丝状、细胞脱落。DHAV-C感染DELC后第12、24、36、48、60、72、84、96小时的病毒量分别为1.23×108、2.14×108、2.95×108、4.37×108、5.75×108、1.70×108、9.55×107和6.46×107 copies/μL。结果表明,本研究成功建立了DHAV-C在DELC上的感染模型,为进一步研究DHAV-C的致病机理、病毒与细胞的相互作用以及建立免疫学诊断方法奠定了基础。The aim of the study was to establish an infection model of DHAV in DELC,to provide a basis for further pathogenesis studies of DHAV-C and virus-cell interaction.In this study,the DELC was prepared with 17-day-old duck embryo and inoculated with DHAV-C.The cytopathic effect(CPE) of DHAV-C to DELC was observed and the dynamic growth curve of DHAV-C in DELC was constructed by real-time reverse-transcriptase PCR.The results showed that CPE first appeared at 36 h post-DHAV-C infection,and the most obvious CPE such as increased cell gap,net-balloon filamentation and exfoliated DELC appeared at 60 h post-DHAV-C infection.The real-time PCR analysis showed the numbers of DHAV-C virus were 1.23×10^8,2.14×10^8,2.95×10^8,4.37×10^8,5.75×10^8,1.70×10^8,9.55×10^7 and 6.46×10^7copies per microliter virus suspension collected at 12,24,36,48,60,72,84 and 96 h post-DHAV-C infection,respectively.The results indicated that we successfully established an infection model of DHAV in DELC which will be useful for further studies on DHAV-C pathogenesis and development of immunological diagnostic methods.
