摘  要:为了评价猪α干扰素-白细胞介素-2重组复合蛋白(rPoIFN-α-linker-PoIL-2)在猪体内抗高致病性猪繁殖与呼吸综合征病毒(HP PRRSV)的效果,采用重组猪α干扰素(rPoIFN-α)-重组猪白细胞介素-2(rPoIL-2)等量混合物、单一rPoIFN-α蛋白、单一rPoIL-2蛋白、rPoIFN-α-linker-PoIL-2复合蛋白等以注射方式进行HP PRRSV的攻毒保护试验,通过分析试验猪体温变化、HP PRRS典型症状维持时间、死亡率、月增重率、病毒血症维持时间等指标,采用RT-PCR和间接ELISA检测攻毒后带毒及抗体产生情况,以评价这些制剂对攻毒猪的保护效果。结果显示,rPoIFN-α-linker-PoIL-2蛋白、rPoIFN-α-rPoIL-2蛋白等量混合物、rPoIFN-α蛋白均可有效抑制HP PRRSV在猪体内的增殖和持续性感染,明显降低发病猪体温,缩短HP PRRS典型症状维持时间,减缓发病症状,降低死亡率;rPoIFN-α-linker-PoIL-2蛋白对HP PRRSV的抑制作用优于单一rPoIFN-α蛋白,稍优于rPoIFN-α-rPoIL-2蛋白等量混合物,显著优于单一rPoIL-2蛋白。上述结果为进一步选取以rPoIFN-α-linker-PoIL-2复合蛋白为主要成分的猪基因工程复合抗病毒制剂研究奠定了基础。To evaluate the antiviral activities of recombinant porcine interferon α-porcine interleukin-2 complex protein(rPoIFN-α-linker-PoIL-2) against high pathogenic porcine reproductive and respiratory syndrome virus(HP PRRSV) in vivo,the pigs were challenged with HP PRRSV at the same times with the equivalent mixture of recombinant porcine alpha interferon protein(rPoIFN-α),and porcine interleukin-2 protein(rPoIL-2),singler PoIFN-α,single rPoIL-2 and rPoIFN-α-linker-PoIL-2 complex protein etc,respectively.The antiviral effect of rPoIFN-α-linker-PoIL-2 complex protein against HP PRRSV in vivo was evaluated by analyzing the indicators of each test group pigs with the changes in body temperature,typical symptoms duration time,mortality rate,the monthly rate of weight gain,viremia duration time etc. The virus shedding and antibody were assessed by PCR and ELISA.The results indicated rPoIFN-α-linker-PoIL-2 complex protein,equivalent mixture of rPoIFN-α protein and rPoIL-2 protein as well as the rPoIFN-α protein could effectively inhibit proliferation and persistent infection of HP PRRSV in pigs,significantly reduce body temperature,shorten the duration of fever and the typical symptoms,and also reduce mortality of the diseased pigs.The antiviral effect of rPoIFN-α-linker-PoIL-2 complex protein on HP PRRSV in vivo was better than single rPoIFN-α protein,slightly better than the equivalent mixture of rPoIFN-α protein and rPoIL-2 protein,and much better than rPoIL-2 protein.This study laid a foundation for further selecting the rPoIFN-α-linker-PoIL-2 complex protein as a main ingredient of porcine gene engineering antiviral agent to prevent and control the HP PRRS and other porcine viral diseases.
