摘 要:随着春保护地西瓜种植面积的扩大和工厂化育苗的发展,西瓜苗期病害也越来越严重,有些常见于生长期的病害,现在在苗期也有较重发生。对湖北省宜城市流水镇育苗场采集的2份西瓜苗期病样,进行分离、纯化并回接病原菌,然后利用PCR扩增真菌保守的ITS序列,初步确定所采集病样1-a的致病菌为枯萎病病原菌[Fusarium oxysporum Schl.f.sp.niveum(E.F.Smith)Snyber et Hansen],病样1-b的致病菌为叶枯病病原菌[Alternaria cucumerina(Ell.et Ev.)Elliott]。本研究利用分子鉴定手段,避免了传统分类方法的繁琐、耗时和结果不稳定性等缺点。With the expansion of planting area and the development of the industrial watermelon seedling production in protected fields, the seedling diseases were becoming more and more seriously, and some common diseases of growing period occurred badly at the seedling stage. In the paper, firstly, we collected two samples with seedling diseases, and separated, purified and inoculated the pathogenic fungi. Secondly, we amplified the conserved ITS sequences of the fungi. Thirdly, we preliminary determinated the pathogenic fungus of sample 1-a and sample 1-b was Fusarium wilt [Fusarium oxyspomm Schl. f. sp. niveum (E. F. Smith) Snyber et Hansen] and leaf blight [Alternaria cucumenna (Ell. et Ev.) Elliott] respectively. In the paper, we used molecular identification method, effectively avoiding the application of tedious, time- consuming and unstable traditional classification methods.
