摘 要:目前,关于microRNA(miRNA)的研究越来越多,但是有关负鼠miRNA的研究却相对较少.为了研究灰色短尾负鼠(Monodelphis domestica)的miRNA,以人和小鼠的miRNA为参考序列,分别利用BLAST、比较基因组学两种方法获得可能的短尾负鼠前体序列(pre-miRNA),并各自用机器学习方法分类鉴定,取这两个结果的交集即得到54条新的短尾负鼠pre-miRNA.将这些新发现的pre-miRNA与miRBase19中已经报道的156条pre-miRNA序列合并,设置miRNA基因间距为5 000 bp时,这些miRNA基因可以形成26个基因簇.这些新发现的miRNA均位于编码基因的间区,同时除部分外,新发现的miRNA基因可以归类到31个已知的miRNA家族.最后,计算分析了保守的新发现的39个miRNA基因的靶基因及其功能.A growing number of researches focus on the microRNA (miRNA) in recent years. In contrast, there remain only a handful of studies on the miRNA of Opossum (Monodelphis domestica). To explore the miRNAs in opossum, candidate precursor miRNAs (pre-miRNAs) have been detected by BLAST search method and comparative genomics approach respectively, and then classified by the support vector machine method. The intersection set of these two results is considered as the new pre-miRNAs and 54 new opossum pre-miRNAs have been discovered. Then these new pre-miRNAs were merged with the 156 pre-miRNA se- quences documented in the miRBase19, and 26 gene clusters were found when the pre-miRNA interval was set as 5 000 bp. Meanwhile, all the 54 new genes was found locating in the inter coding genic region and they could be classified into 31 known gene families. Finally, the molecular functions of the target genes of the 39 conservative and newly discovered pre-miRNAs in opossum have been computational analyzed.
