摘  要:用铝塑泡罩包装板在即将凝固的不同浓度琼脂糖(2%、3%、4%、5%)溶胶板上压制小孔,用作胚胎免疫染色操作载体,比较以琼脂糖凝胶孔为载体和以一次性细胞培养皿为载体进行胚胎免疫染色的效果。结果表明,以琼脂糖凝胶孔作为载体进行胚胎免疫荧光染色与传统载体染色效果相同,但避免了操作过程中由于胚胎与载体的黏附而造成的诸多不便;以3%琼脂糖凝胶孔为载体孔内液体损失率最小;琼脂糖凝胶板在4℃保存至少两周不影响其使用效果,并可反复使用。琼脂糖凝胶孔较传统载体更适于进行胚胎免疫荧光染色。The traditional medium for embryo immunofluorescence staining is a disposable culture dish, embryo adhesion-prone, which is unfavorable for the following researches. In this study, instead of the dish, agarose was used at different concentrations (2% , 3% , 4% and 5% ) for embryo immunofluorescence staining by pressing a aluminiumplastic blister package onto it. The results ofimmunofluorescence staining using two different medium (agarose gel hole and tissue culture plate) were compared. The staining effect of embryo in agarose gel hole was the same as that stained in disposable culture dish, but the new medium could avoid embryo adhesion. The minimum loss of liquid was in 3% agarose gel hole, which was the most suitable as medium for immunofluorescence staining. The gel plate could be reused after stored at 4 ℃for at least two weeks. It indicated that agarose gel hole might be more suitable for embryo immunoflurorescence staining than the diposable dish.
