摘 要:本研究采用表面消毒后涂布平板的方法,从开封郊区不同品种的45份健康小麦根系内分离获得105株小麦内生细菌.采用种子包衣法,在小麦活体上测定了内生细菌对小麦黄斑叶枯病的生防效果.其中,菌株T-7生防效果最好,防治效果为53.41%.综合利用形态学、生理生化分析等方法对T-7进行鉴定,结果表明,T-7属于枯草芽孢杆菌(Bacillus subtilis).定殖能力测试表明,T-7在小麦根部可以长期定殖.处理30天后,该菌株在小麦根部的菌落形成单位仍达到3.4×104 CFU/g roots.A total of 105 wheat endophytic bacterias were isolated from 45 healthy root systems of wheat varieties originated from Kaifeng suburb by the methods of surface disinfection and spread plate.By seed-treatment with bacterial fermentation filtrate,in vivo tests were used to acquire an efficient biocontrol strain against wheat leaf blotch.The results showed that T-7 strain could significantly reduce the disease severity of wheat leaf blotch.Control efficacy of strain T-7 against Rhizocotonia sonali reached 53.41%.Methods of morphological observation,physiological and biochemical analyses were used to identify the biocontrol strain T-7.The results indicated that T-7 strain belongs to Bacillus subtilis.The colonization assay indicated that the T-7 strain could persistently survived on the wheat roots and has a significantly ability of colonization.The concentration of T-7 strain was 3.4×104 CFU/g roots in the wheats after 30 days of inoculation.
