摘  要:为了解宁夏固原地区马铃薯M病毒(potato virus M,PVM)发生情况,采集了18份表现叶片变形、茎坏死及植株矮化症状的样品,应用血清学(DAS-ELISA)和分子生物学(RT-PCR)方法进行检测。DAS-ELISA检测结果显示,有2份样品(5号和10号)为PVM阳性。对2份阳性样品进行RT-PCR检测,均获得416bp的目的产物,其序列与国内外已报道的其他PVM分离物的核苷酸序列同源性最高分别达99%和98%,说明被检测样品确实感染了PVM。这是PVM在宁夏固原发生的首次报道,为今后该地区马铃薯病毒病害的防控提供了依据。In order to know the occurrence of potato virus M (PVM) in Guyuan of Ningxia,18 samples displaying symptoms of leaf distortion,stem necrosis,and plant dwarf were collected and tested.Enzyme-linked immunosorbent assay (ELISA) showed positive reaction of PVM in two samples (No.5 and No.10).The two samples were detected by RT-PCR and a target band with size about 416 bp was amplified from both samples.The sequence analysis of amplified fragment showed that the highest homology of the two sequences with other isolates of PVM was 99% and 98%,respectively,which further indicated that the samples were infected by PVM.It was the first report of PVM occurrence in Guyuan of Ninxia,which provided a basis for prevention and control of potato virus disease in this region in the future.
