地塞米松与IL-10联合作用对脓毒症大鼠L6肌管细胞蛋白代谢的影响

摘　　要：目的探讨地塞米松(dexamethasone,DEX)与IL-10联合作用对体外培养的脓毒症大鼠血清处理的L6肌管细胞蛋白代谢的影响。方法将分化的大鼠L6肌管细胞分A、B、C组,A、B两组均以脓毒症大鼠血清处理12h,C组细胞以PBS处理作为空白对照组。A组细胞以DEX作用于L6细胞24、48h,B组细胞以DEX+IL-10作用于L6细胞24、48h,C组细胞以正常培养液作为对照组。荧光实时定量PCR检测糖皮质激素受体(glucocorticoid receptor,GR)的mR-NA表达水平;Western blot检测GR、肌球蛋白重链(myosin heavy chain,MHC)的蛋白表达水平。结果 B组GR的mRNA及蛋白表达均较C组和A组明显降低(均P<0.05),而B组MHC蛋白表达均较C组和A组明显升高(均P<0.05),GR蛋白表达与MHC的蛋白表达有明显的负相关性(均P<0.05)。结论 IL-10可降低肌管细胞GR的表达并减少MHC的降解,IL-10对脓毒症大鼠骨骼肌的蛋白代谢起到一定的正向调节作用。Objective To explore the effects of combined use of dexamethasone（DEX）and IL-10on the protein metabolism of L6myotube cells treated with sepsis rat serum.Methods The differentiated rat L6cells were divided into A,B,C groups.Cells in both A and B groups were treated with sepsis rat serum for 12h,while those in group C were treated with PBS and served as blank control.Additionally,L6cells in group A were treated with DEX for 24and 48h,and those in group B with DEX＋IL-10for 24and 48hand those in group C with normal medium for the same time.The mRNA expression level of glucocorticoid receptor（GR）was determined by fluorescence real time quantitive PCR.The protein expression level of GR and myosin heavy chain（MHC）was detected by Western blot.Results The mRNA and protein expression levels of GR were decreased obviously in group B compared with group C and group A（P〈0.05for all）.The protein expression level of MHC was increased evidently in group B compared with group C and group A（P〈0.05for all）.The protein expression of GR was negatively correlated with that of MHC（P〈0.05）.Conclusion IL-10can decrease the expression of GR and increase MHC degradation,which may positively regulate the protein metabolism of skeletal muscle in sepsis rats.
