摘 要:目的研究原花青素(Procyanidin,PC)对氟致小鼠成釉细胞DNA损伤的拮抗作用。方法选择体外培养的小鼠成釉细胞,分别设立对照组;0.25、0.50、1.00、2.00、4.00mmol/L氟化钠(NaF)单独染毒组;10.00、25.00、50.00mg/L PC单独染毒组以及10.00、25.00、50.00mg/L PC+0.25、0.50、1.00、2.00、4.00mmol/L NaF联合作用组。细胞作用24h后,采用单细胞凝胶电泳技术(SCGE)检测DNA损伤情况。结果与对照组比较,NaF单独染毒组小鼠成釉细胞的DNA尾长、Olive尾矩、尾DNA%、尾长/头长值显著性升高(均P<0.05),而PC单独染毒组小鼠成釉细胞DNA损伤的上述各项指标显著下降(均P<0.05)。10.00mg/L PC+0.25、0.50、1.00、2.00mmol/L NaF联合作用组小鼠成釉细胞DNA损伤的各指标和25.00、50.00mg/L PC+各剂量NaF联合作用组小鼠成釉细胞的DNA尾长、尾长/头长值明显低于对照组和相同剂量NaF单独染毒组(均P<0.05)。25.00mg/L PC和50.00mg/L PC+各剂量NaF联合作用组小鼠成釉细胞的Olive尾矩明显高于10.00mg/L PC组(均P<0.05)。结论过量氟可致小鼠成釉细胞DNA损伤,而适量原花青素对氟致小鼠成釉细胞DNA损伤有拮抗作用,且其浓度为10.00mg/mL时作用效果较为明显。Objective To investigate the antagonistic effects of procyanidin(PC)on fluoride-caused DNA damage in mouse ameloblasts.Methods The mouse ameloblasts were cultured in vitro and treated with different concentrations of sodium fluoride(NaF 0.25,0.50,1.00,2.00or 4.00mmol/L),or/and different concentrations of PC(10.00,25.00or 50.00mg/L)for 24 h.The single cell gel electrophoresis was used to measure the DNA damage in mouse ameloblasts.Results The tail length,olive tail moment,DNA%and tail/head length ratio were significantly increased in mouse ameloblasts in NaF groups compared with control group(P〈0.05).And these DNA damage indices were significantly improved after PC treatment(P〈0.05).The DNA damage indices in PC(10.00mg/L)+NaF(0.25-2.00mmol/L)groups and the tail length,tail/head length ratio in PC(25.00,50.00mg/L)+NaF groups in mouse ameloblasts were much lower than in the control group and the corresponding NaF groups(P〈0.05).Compared with PC(10.00mg/L)+NaF groups,the olive tail moment in PC(25.00,50.00mg/L)+ NaF groups were substantially increased(P〈0.05).Conclusion Excessive fluoride can induce DNA damage in mouse ameloblasts,which can be antagonized by PC,and PC at the dose of 10.00mg/L has optimal effect.
