摘 要:以黄瓜品种长春密刺子叶节为外植体,研究了乙酰丁香酮(AS)的添加方式、浓度及共培养pH对黄瓜遗传转化的影响。结果表明:在侵染菌液和共培养基中加入AS均可显著提高黄瓜抗性芽诱导率,浓度以100μmol·L-1为最佳。共培养基的pH对黄瓜抗性芽诱导率也有影响,其中在pH 5.2的共培养基上黄瓜转化率最高。研究结果优化了黄瓜遗传转化体系,提高了黄瓜转化效率,为推动黄瓜转基因工作奠定了坚实的基础。Cucumber genetic transformation affected by acetosyringone (AS) concentration applied, adding mode and coculture pH was studied with cotyledonarynode of cucumber "Changchun mici". The results indicated that both AS added in infection bacteria liquid and co-culture medium could significantly improve the rate of resistance bud induction. The optimum AS concentration was 100 p, mol/L. Cucumber resistance bud induction rate was also affected by different pH of co-cul- ture medium and the optimum pH for cucumber genetic transformation was 5.2. These research results were helpful to optimize system of cucumber genetic transformation and transgenic research of cucumber.
