摘  要:旨在研究四川中江丹参轮作期间土壤真菌遗传多样性变化特征。分离提取土壤微生物DNA,PCR特异扩增,以DGGE分离扩增片段并测序,在GenBank中查得菌种。结果表明:中江丹参轮作期间,土壤真菌群落包括:Termitaria sp.YAKU2,Agaricus bisporus,Cercozoan,Cordyceps,Geomyces destructans,Geomyces sp.P7,Cordyceps brongniartii,Penicillium sp.KCTC6774,uncultured Iodophanus clone S-54。聚类分析表明,休地1年与正茬丹参的土壤真菌群落结构相似,而与休地3、4年的土壤真菌群落有明显差异;休地2年是属于过渡状态。研究认为中江丹参轮作期间,土壤真菌群落遗传多样性结构逐渐恢复;而且休地的第2年是土壤真菌群落结构恢复的关键时期,与前期报告结论一致。In this paper, the aims were to study genetic diversity variation characteristics of soil fungal during Salvia Miltiorrhiza Bge. cropping rotation period in Zhongjiang County of Sichuan Province. Soil microbial DNA was isolated and extracted; then specific DNA fragments of fungi were amplified by PCR; the amplifying DNA fragments were separated by DGGE. The strains were gained in GenBank by DNA sequencing. The result showed that during the Salvia Miltiorrhiza Bge. cropping rotation period in Zhongjiang County, soil fungal community included: Termitaria sp. YAKU2, Agaricus bisporus, cercozoan, Cordyceps, Geomyces destructans, Geomyces sp. P7, Cordyceps brongniartii, Penicillium sp. KCTC6774, uncultured Iodophanus clone S-54. Cluster analysis showed that the soil fungal community was similar between fallow land 1 year and this year; but it was obviously different from that of fallow land 3 years and 4 years. It was geared to the style of transition about fallow land 2 years. It was concluded that during the Salvia Miltiorrhiza Bge. cropping rotation period in Zhongjiang County, the soil fungal community and genetic diversity gradually recovered; the second year offallow land was the key period of soil fungal community to restore; it was in agreement on the previous report conclusion.
