摘  要:目的:观察钾离子通道开放剂尼可地尔和阻滞剂格列苯脲对哮喘大鼠肺泡盥洗液(BALF)中IL-4/IFN-γ的影响以及对气道重塑的作用。方法:SD雄性大鼠50只,随机分为正常对照组、哮喘模型组、尼可地尔组、地塞米松组和格列苯脲组。采用卵清蛋白(OVA)致敏复制大鼠哮喘模型,模型成功后取相关组织分别进行图像分析测定各组支气管壁各层的厚度、ELISA检测肺泡盥洗液(BALF)中IL-4/IFN-γ表达情况。结果:哮喘模型组和格列苯脲组支气管平滑肌明显增厚(P<0.05),有典型的气道重塑表现,尼可地尔组和地塞米松组无明显增厚;尼可地尔组和地塞米松组IL-4表达低于哮喘模型组和格列苯脲组,IFN-γ表达高于哮喘模型组和格列苯脲组(P<0.05)。结论:尼可地尔通过开放KATP通道,能够明显改善哮喘大鼠气道炎症和抑制哮喘大鼠气道重塑的形成,其可能机制与IL-4表达降低,同时IFN-γ表达升高有关。Objective:To observe the effects of Glibenclamide and Nicorandil on the level of IL-4/IFN-γin BALF and airway remodeling in asthma rats.Method:50 SD male rats were randomly divided into five groups:the normal control group,the asthma model group,the nicorandil group,the hexadecadrol group and the glibenclamide group.The asthma model was established by ovalbumin sensitizing-challenging rats.The thickness of bronchial wall was detected by computer image analyse system,the IL-4 and IFN-γin BALF were detected by ELISA.Result:The thickness of airway smooth muscle was increased in the asthma model group and the Glibenclamide group( P 0.05),which showed pathological changes of airway remodeling.It was not obvious in the Nicorandil group and the hexadecadrol group.The content of IL-4 of BALF in the Nicorandil group and the hexadecadrol group was lower than that in the asthma model group and the Glibenclamide group,but the level of IFN-γwas higher( P 0.05). Conclusion:Nicorandil could obviously alleviate the airway inflammation and inhibit the formation of the airway remodeling,which may account for the opening of ATP-sensitive potassium channel in asthma rats.The possible mechanism might be related to the reduction of expression of IL-4 and the rise of the expression of IFN-γ.
