行气调神针刺疗法对缺血性脑卒中后抑郁模型大鼠脑左前皮质及海马cAMP含量、PKA和PKC活性的影响

摘　　要：目的探讨行气调神针刺疗法对缺血性脑卒中后抑郁(PSD)模型大鼠脑左前皮质及海马环磷酸腺苷(cAMP)、蛋白激酶A(PKA)和蛋白激酶C(PKC)的影响,初步阐明针刺治疗PSD的作用机制,为针刺治疗PSD提供实验依据。方法通过电凝大鼠大脑中动脉引起局部脑缺血,并合并应激及孤养致PSD模型,以行气调神针刺疗法及氟西汀灌胃治疗6周,采用生物素双抗体夹心酶联免疫吸附法(ELISA法)检测缺血性PSD大鼠脑左前皮质及左侧海马中的cAMP含量、PKA和PKC活性的变化。结果模型组大鼠脑左前皮质及海马cAMP含量及PKA活性与正常组比较显著降低,PKC活性显著升高,差异有高度统计意义(P<0.01);针刺组、西药组cAMP含量及PKA活性较模型组显著升高,PKC活性显著降低,2组比较差异有高度统计意义(P<0.01);针刺组与西药组比较差异无统计意义(P>0.05)。结论行气调神针刺疗法的抗抑郁作用与氟西汀相当,其作用机制可能与调节cAMP及磷酸酰肌醇(PI)信号通路有关。Objective To explore the effects of Xingqi Tiaoshen acupuncture therapy on cyclic adenosine monophosphate （ cAMP ） content, protein kinase A （PKA） and protein kinase C （ PKC ） activity of left anterior cortex and hippocampus in ischemic post-stroke depression（PSD）rats, clarify the mechanism of acupuncture treating PSD, and provide experimental basis for treating PSD by acupuncture. Methods By electric coagulation of middle cere- bral artery in rats induced cerebral ischemia, and complicated with stress to induce PSD model. The rats had been treated for 6 weeks by Xingqi Tiaoshen acupuncture therapy and fluoxetine gavage. The cAMP content, PKA and PKC activity of left anterior cortex and hippocampus in ischemic PSD rats were detected by ELISA method. Results Compared with normal group, cAMP content, PKA activity of left anterior cortex and hippocampus of model group rats were significantly decreased, and PKC activity was significantly increased（P〈0. 01 ）. Compared with model group, cAMP content and PKA activity of acupuncture group and western medicine group were significantly in- creased, PKC activity was significantly decreased（P〈0.01 ）. The difference between the acupuncture group and western medicine group hadn＇t statistics significance（P〉0.05）. Conclusion The antidepressant effect of Xingqi Tiaoshen acupuncture therapy is equal to that of fluoxetine. The mechanism can related to adjust cAMP and PI sig- naling pathways.
