摘　　要：目的　研究蝮蛇毒蛋白 C激活物的分离纯化与理化性质。方法　经 DE5 2 -纤维素、CM-Sphadex C-5 0、G-75柱层析 ,从安徽芜湖产蝮蛇 ( Agkistrodon halys)蛇毒中纯化一种均一的蛋白 C激活物 ( PCA)。结果　SDS-PAGE测定分子量约为 1 5 5 0 0 Da,IEF-PAGE测定等电点为 4 .8。它能使人血浆的 KPTT明显延长 ,显示出强烈的抗凝活性。通过中和试验与显色肽定量实验表明 ,它能特异性激活蛋白 C,并且激活作用与剂量呈线性关系。s Objective To study the method of purifying protein C activator of Agkistrodon halys pallas and physical and chemical character Methods A protease from venom of Wannan Agkistrodon halys pallas was purified to homogeneity by a combination of DEAE celluose、CM sephadex C 50、Sephadex G 75 Results The molecular weight was estimated to be 15 5KD by SDS PAGE and isoelectric point determined by isoelectric focusing polyacrylmide gel electrophoresis was 4 8 It was able to markedly prolong the clotting time of human plasma and showed the strong anticoagulant activity The purified enzyme was proved to be protein C activator by neutralization and amidolytic assay of protein C
