摘 要:目的探讨HBV对人骨髓来源间充质干细胞(MSCs)体外增殖的影响。方法从成人骨髓分离纯化MSCs并体外培养至第3代,将MSCs与含高浓度HBV的人血清共培养作为实验组,并用不含HBV的健康人血清作为对照。采用MTT法绘制细胞生长曲线并计算细胞倍增时间。分别应用细胞克隆形成试验和流式细胞术检测MSCs的克隆形成能力和细胞增殖指数。结果实验组的生长曲线发生右移,其对数生长期的细胞倍增时间较对照组延长(t=2.453,P<0.05)。体外培养10 d后,实验组的细胞克隆形成率和增殖指数均明显低于对照组(t=2.488,P<0.05;t=2.798,P<0.05)。结论 HBV可抑制人骨髓间充质干细胞的体外增殖。Objective To investigate the effect of HBV on proliferative capacity of human bone marrow derived mesenchymal stem cells (MSCs) in vitro. Methods MSCs were isolated from human bone marrow and cultured for the third passage. They were divided into two groups: some were cocultured with HBV- containing serum as experimental group, and the others were cocultured with healthy serum as control. Cell growth curve and cell doubling time were evaluated by MTT assay. Colony-forming capacity and cell cycle were evaluated by colony-forming assay and flow cytometry. Results In contrast, growth curve turned right and cell doubling time significantly increased (t = 2.453, P 〈 0.05). After 10 days culture, colony-forming rate and proliferative index in experimental group were significantly lower than those in control group (t = 2.488, P 〈 0.05; t = 2.798, P 〈 0.05). Conclusion HBV could inhibit proliferative capacity of human bone marrow derived MSCs in vitro.
