摘  要:目的:观察α-胞衬蛋白(α-Fodrin)的小干扰RNA(small interfering RNA,siRNA)对干燥综合征动物模型非肥胖糖尿病(NOD)小鼠细胞因子IL-2、IL-6和TGF-β的影响及泪腺的病理改变。方法:0.5 mg/kg体重的pGFPV-RS空载体和α-Fodrin-siRNA表达载体分别尾静脉注射NOD小鼠,1周/次,共2次。注射后2、4、6 d尾部采血,酶联免疫吸附试验(Enzyme linked immunosorbent assay,ELISA)检测血清中细胞因子IL-2、IL-6和TGF-β的表达;注射后7 d病理切片,苏木精-伊红(HE)染色法观察各组小鼠泪腺的病理变化。结果:注射后2 d和4 d siRNA 1组和siRNA2组NOD小鼠血清中IL-6的浓度明显低于对照组和空载体组,差异有统计学意义;而对照组和空载体组间、siRNA 1组和siRNA 2组间比较差异无统计学意义。注射后2、4和6 d siRNA 1组和siRNA 2组NOD小鼠血清中TGF-β和IL-2的浓度较对照组和空载体组升高,但差异无统计学意义。病理切片结果显示,siRNA 1组和siRNA 2组与对照组和空载体组比较,小鼠泪腺淋巴细胞浸润减少。结论:NOD小鼠注射α-Fodrin siRNA载体后,血清中IL-6的水平降低、IL-2和TGF-β的浓度升高,泪腺淋巴细胞浸润减少,提示α-Fodrin siRNA可以减轻炎症反应。Objective: To observe effect of small interfering RNA (siRNA) of α- Fodrin on IL - 2, IL - 6 and TGF -β in serum of NOD mice and the pathological changes in their lacrimal glands. Methods: pGFP - V - RS empty vectors and α-Fodrin -siRNA expression vectors (0.5 mg/kg) were injected by tail veil into NOD mice, once a week and two times totally. Blood was taken from the tail veil on the 2nd, 4th and the 6th day after the injection and the expression level of IL -2, IL -6 and TGF -β in serum was assayed by enzyme linked immunosorbent assay (ELISA). Pathological section was made on the 7th day after the injection and pathological changes of tbe lacrimal gland in the mice from different groups were observed with hematoxylin - eo- sin (HE) staining. Results: The level of IL -6 in sermn of NOD mice was significantly lower in siRNA1 group and siRNA2 group than that in the control and empty vector group on the 2nd and 4th day after the injection ( P 〈 0.05). There was no sig- nificant difference in the level of IL- 6 between the control group mid the empty vector group, and between siRNA1 group and siRNA2 group, on the 2nd and 4th day after the injection. The level of TGF -β and IL -2 was higher in siRNA1 group and siR- NA2 group than that in the control group and empty vector group on the 2nd and 4th and 6th day after the injection, without sta- tistical significance ( P 〉 0. 05). Compared with the control group and empty vector group, the lymphocyte infiltration of lacri- ntal gland was significantly reduced in siRNAlgroup and siRNA2 group. Conclusion: After the injection of α- Fodrin specific siRNA, the level of IL - 6 in the serum drops and the level of TGF -β and IL - 2 rises, suggesting that α- Fodrin specific siR- NA can reduce inflammatory reaction.
