人红细胞非凝集型单抗(自身全血凝集试验试剂)的研制及鉴定

摘　　要：应用杂交瘤技术 ,混合人O型红细胞免疫纯系BALB/C小鼠 ,包被的红细胞作为固相抗原 ,ELISA法筛选出识别红细胞表面共同抗原的抗体 ,然后利用Coombs原理 ,加入抗球蛋白试剂。用玻片法筛选出非凝集型抗体 ,得到 2株稳定分泌上述抗体的杂交瘤并进行初步鉴定 ,单抗上清血凝效价可达 1∶5 0～ 1∶10 0 ,腹水血凝效价为 1∶8× 10 4 ～ 1∶16× 10 4 。另外 ,本文特别讨论了凝集型单抗的Fab段作为非凝集型抗体应用的可能性。The spleen cells of BALB/C mice immunized with pooled human type 0 erythrocytes were hybridized with SP 2/0 myeloma cells.Using pooled human erythrocytes(A,B,AB,0,coated respectively)as solid antigen,the antibodies to common surface antigen of human erythrocytes were screened out by RBC ELISA.According to the Coombs principle,by adding anti immunogobulin into the mixture of erythrocytes and McAbs,the non agglutinating antibodies were selected.Using limited dilution four times for cloning and subcloning,two strains of hybridoma cells which secrete non agglutination antibodies were obtained.Their antibodies have hemagglutination titers of 1∶50 100 and 1∶8 16×10 4 for supernatant and ascite,respectively.Besides,the possibility of using the Fab fragments of agglutinating McAbs as non agglutinating antibodies was especially discussed.
