摘 要:目的验证补肾类中药有效成份是否具有促进体外培养成骨细胞增殖的作用。方法选用仙茅、菟丝子、沙苑子的黄酮和总多糖成份,用MTT比色法测定各药物组活细胞吸光度值(optical density,OD),计算出不同浓度药物对细胞生长的抑制率,并对实验数据进行统计分析,比较细胞毒性大小。结果三种药物的黄酮提取成份均有较强的细胞毒作用;总多糖成份在合适的浓度下均能促进成骨细胞增殖。结论本实验从细胞水平证明了"肾主骨理论"的正确性,并推断补肾药促进成骨作用的主要成份是总多糖。Objective To verify the effective? components of kidney-reinforcing drugs for promoting osteoblasts proliferation in vitro. Methods Effects of flavones and total polysaccharide of Rhizoma Curculiginis, Semen Cuscutae and Semen Astragal Com- planati on osteoblasts proliferation were tested by using a MTT colorimetric method. Inhibition rates of cell growth by different concentra- tions of each component were calculated, and then, cytotoxic effects of each component were compared. Results Three kinds of flavones components had a strong cytotoxicity while proper concentrations of three kinds of total polysaccharide components could promote prolif- eration of cuhured osteoblasts. Conclusion The experiments demonstrate that the TCM theory of "kidney governing bones" is true at cell level, and deduce that the effective? components of kidney-reinforcing drugs for promoting osteoblasts proliferation may be total polysaccharide.
