摘 要:为研究不同毒力的猪肺炎支原体对宿主细胞的黏附作用,本研究采用体外培养的猪气管环检测不同毒力的猪肺炎支原体代表菌株对猪气管上皮细胞及纤毛的损伤情况,初步验证不同猪肺炎支原体代表菌株的毒力。应用间接免疫荧光技术研究不同猪肺炎支原体分离株、标准株、168致弱株及疫苗株对猪肾细胞PK15、肺泡上皮细胞SJPL和猪肺泡巨嗜细胞3D4/31的黏附情况。结果表明猪肺炎支原体野毒株XLW-1能引起猪气管上皮细胞严重病变及纤毛损伤、脱落;国际标准弱毒株J株仅能引起猪气管上皮细胞轻微病变;168疫苗株既不能引起猪气管上皮细胞病变,也不能引起纤毛损伤。野毒株XLW-1、国际标准强毒株232株、弱毒株J株及168疫苗株均能黏附PK15、SJPL和3D4/31,168致弱株可以黏附PK15和SJPL。野毒株、标准强毒株、标准弱毒株、168致弱株及疫苗株均能黏附PK15、SJPL和3D4/31。he purpose of this study is to investigate the adhesion to host cells of different virulent Mycoplasma hyopneumoniae. The virulence of different virulent M.hyopneumoniae representative strains would be verified by damage of swine tracheal epithelial cells and ciliary via porcine tracheal rings cultivated in vitro. The adhesion of different M.hyopneumoniae isolates, standard strains, attenuated strain 168 (more than 100-passage) and vaccine strain 168 (more than 300-passage) to porcine kidney cell (PK15), lung epithelial cells (SJPL) and porcine lung macrophage (3D4/31) was further investigateed by detection with indirect immunofluorescence assay. The results showed that M.hyopneumoniae wild strain XLW-1 was able to induce obvious epithelium cytopathic effect (ECE), cilia damage and shed. International standard low virulent strain J only induced slight cytopathy in porcine tracheal epithelial cells. However, vaccine strain 168 neither induced swine tracheal ECE, nor cilia damage. Moreover, wild strain XLW-1, international standard virulent strain 232, international standard low virulent strain J and vaccine strain 168 all were able to adhere to PK15, SJPL and 3D4/31. The attenuated strain 168 only sticked to PK15 and SJPL. While, all of wild strain, international standard virulent and low virulent strain, attenuated strain 168 and vaccine strain 168 were able to adhere to PK15, SJPL and 3D4/31.
