摘  要:目的探讨L-选择素基因单核苷酸多态性与系统性红斑狼疮(SLE)易感性之间的关系;同时分析L-选择素基因型及血清水平与SLE的相关性。方法以145例SLE患者和160例健康对照者为研究对象,应用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)和DNA测序的方法对L-选择素基因P213S单核苷酸多态性进行基因分型,同时采用酶联免疫吸附试验检测血清L-选择素水平。结果在SLE组中,L-选择素三种基因型PP,PS,SS分布频率分别为54.48%,38.62%和6.90%;在对照组中其分别为51.88%,40.00%和8.13%。L-选择素基因P213S多态性在SLE组和正常人群中的分布频率差异无统计学意义(P>0.05),但携带PP基因型的SLE患者血清L-选择素水平显著高于不携带者,差异有统计学意义(P<0.05)。SLE组血清L-选择素水平明显高于对照组,差异有统计学意义(P<0.05)。结论 L-选择素基因P213S多态性与SLE的易感性无关,但PP基因型携带者可能与SLE的发病风险相关。Objective To investigate of the single nucleotide polymorphism of L-selectin gene are associated with susceptibility of systematic lupus erythematosus (SLE) in a Chinese population, and to analyze the association of serum levels and genotypes of L-selectin with SLE. Methods We analyzed L-selectin gene P213S single nucleotide polymorphism in 145 patients with SLE and 160 age and sex matched controls in a Chinese population,using a polymerase chain reaction - restriction fragment length polymorphism (PCR-RFLP) strategy and DNA sequencing, and the serum levels of L-selectin was determined by enzyme-linked immunosorbent assay (ELISA). Results The frequencies of PP,PS and SS genotype were 54.48% ,38.62% and 6.90% in SLE patients respectively, while in controls were 51.88% ,40.00%, and 8.13%. The distributions of L- selectin gene P213S polymorphism was not significantly different between SLE and control group (P 〉 0.05 ), the serum levels of L-selectin PP genotype carriers was significantly higher than that of no carriers ( P 〈 0.05). L-selectin serum levels of the patients with SLE were significantly higher than those of the controls (P 〈 0.05 ). Conclusion L-selectin gene P213S polymorphism are not associated with susceptibility of SLE, Carrying L-selectin PP genotype increase the risk of SLE.
