摘  要:本文报道了二齿新蚤和宽新蚤线粒体 DN A中一段 90 0碱基的片段 ,其中包括完整的 COII基因和两个氨基酸 t RNA基因片段。两种新蚤 COII基因的 CG含量都是 33.3% ,这在昆虫中是较高的。两者基因间碱基的变异率为 1.8% ,变异以 T→ C转换为主 ,显示两种新蚤间较近的亲缘关系。两种新蚤氨基酸 t RNA基因碱基的变异率低于 0 .5%。两种新蚤的 COII基因和猫栉首蚤 COII基因间碱基的变异率大于 2 0 %。同时列出了完整的细胞色素氧化酶亚基 II氨基酸序列。并列出了在这个基因片段中一些保守序列的位置。A fragment of mtDNA is cloned and sequeenced from Neopsylla bidentatiformis and Neopsylla mana, which includes a complete COII gene and flanking partial Leu tRNA gene and Lys tRNA gene.The CG content of COII gene is 33 3% for two species,which is higher in insect.The sequence difference of COII gene between the two species is 1 8% and is caused mainly by the transitions of T→C.It is suggested that these two species have an intimate relationship.The difference of two tRNA genes is lower than 0 5%.The amino acid sequence difference of COII between the two Neopsylla species is 2%.The sequence difference of COII gene among Neopsylla and Ctenocephalides felis is more than 20%.In addition,some of the conservative sequences in COII gene are labled.
