摘  要:从2130尾性成熟的雄性罗氏沼虾体内采集雄性腺体(AG),匀浆后,分离沉淀等电点为4.5左右的多肽,用十二烷基磺酸钠-聚丙烯酸胺凝胶电泳法收集分子量从17kDa至18kDa的多肽条带。进一步应用高效液相色谱、无胶筛分高效毛细管电泳、基质辅助激光解吸电离飞行时间质谱、高效液相色谱-电喷雾电离-质谱等技术进行分离与鉴定。结果显示,主要组分的分子量为17436±110u,在等电点4.5的条件下分离得到的罗氏沼虾AG组分中,以分子量17480u的多肽组分为主,与等足类ArmadillidiumvulgarsAGH的分子量相似,暂称为等足目AGH类似物。The androgenic gland was collected from male Macrobrachium rosenbergii,then was homogenized with 0.1mol / L NaAc soluhon. The homogenate was centrifgated 15min at 12000 r /min, the upper layer clear solution was adjusted to pH4.5,then was centrifugated again and the precipitate was collected. This precipitate was separated with SDS-PAGE method. The crude product of the isopada AGH analogue (Androgenic Gand Hormone) of M. rosenbergii was collected from the protein belt which molecular weight range from 17k Da to 18k De by SDS-PAGE method. The applications of new techniques such as HPLC, and MALDL-TOF-MS were carried out.Matrix-assisted laser desorption ionization time-of -flight mass spectrometry and HPLC-ESI-MS methods may provide a more sensitive and accurate determination for the molecular weight of AGH. The molecular weight of the isopada AGH analogue is 17480u as identified by HPLC-ESI-MS and 17436±110u by capillary electrophoresis.
