鸡MD肿瘤细胞DNA含量及细胞凋亡的特性

摘　　要：用SPF来杭鸡复制MD肿瘤模型 ,取MD肿瘤组织和相同日龄鸡的正常组织 ,制成单个细胞悬液 ,碘化丙锭染色后 ,用流式细胞仪检测细胞DNA含量和细胞凋亡率。结果 :(1)MD肿瘤有二倍体肿瘤和异倍体肿瘤 ,31例MD肿瘤中2 6例为二倍体肿瘤 ,5例是异倍体肿瘤 ;(2 )MD肿瘤细胞动力学特点是 :DNA合成期 (S期 )细胞比例明显增加 ,肾、肝、卵巢和睾丸MD肿瘤细胞S期分别为 12 13 % ,11 30 % ,12 31%和 15 2 6 % ,相应组织细胞依次为 1 37% ,1 2 7% ,1 5 4%和 0 0 7% ,增殖指数 (proliferationindex ,PI)显著增高 ;(3)MD肿瘤细胞凋亡率明显低于正常组织 ,肾、肝、卵巢和睾丸MD肿瘤细胞凋亡率分别是 0 33% ,0 6 9% ,0 73 % ,0 16 % ,而相应组织细胞凋亡率则为 1 40 % ,2 44 % ,1 92 %和 2 11%。提示细胞周期紊乱、细胞增殖活跃。MD tumor model was reproduced in SPF White Leghorn chickens.MD tumor and normal tissues of chicken in same day old were collected and suspended in PBS with the single cell.The DNA content was measured and apoptotic rate was calculated with flow cytometry after being stained with propidium iodide.The results showed that:(1)There were two types tumor in MD:diploid and aneuploid tumor.26 of 31 are diploid tumor,and the others were aneuploid tumor;(2)Characteristics of cell cycle on MD tumor:cell ratio of S phase(DNA synthetic phase)raised remarkably on kidney,liver,ovary and testis which was 12 13%,11 30%,12 31% and 15 26% respectively and in relevant normal tissue,the value was 1 37%,1 27%,1 54% and 0 07% respectively.Proliferation index(PI)increased obviously;(3)Apoptotic rate of MD tumor cell was much lower than that of relevant normal tissue cell.Apoptotic rate of MD tumor on kidney,liver,ovary and testis was 0 33%,0 69%,0 73% and 0 16% respectively,but that of normal tissues was 1 40%,2 44%,1 92% and 2 11%.It is indicated that cell cycle disorder,cell proliferation activity and apoptosis inhibition are the cytology base in lyphomagenesis of MD.
