摘  要:目的建立并完善贵州省禽流感病毒基因分析检测技术平台,了解贵州省禽流感病毒H5N1亚型HA1片断与WHO公布的流行株比对有否发生变异。方法收集禽流感H5N1亚型病毒毒株,提取病毒核酸RNA,通过RT-PCR扩增HA1片断,用电泳观察PCR产物目的条带大小,经过纯化、测序,用Biodeit、MEGA4相关软件进行序列比对,进行差异性分析,构建系统发育树。结果贵州省禽流感病毒H5N1亚型中鸭来源的禽流感病毒组成一个分支,组内差异性在0.00~5.00之间;鸡来源的禽流感病毒组成另外一个分支,组内高度同源,差异性为零。结论贵州省禽流感病毒在物种之间存在一定的种属特异性。并且与湖南省的禽流感病毒亲缘关系较近,这与两省之间地理交界有关。Objective To establish and improve the technology platform of gene analysis and de- tection of avian influenza virus in Guizhou province, and to know whether the epidemic strain compari- son in Guizhou province HSN1 subtype avian influenza virus HA1 fragment and WHO announced a change. Methods Collection of H5N1 subtype avian influenza virus strains, extraction of virus nucleic acid RNA. HA1 fragment was amplified by RT-PCR, using electrophoresis were used to observe the PCR product band size, after purification, sequencing, Biodeit, MEGA4 related software for sequence alignment, analyze the differences, to construct a phylogenetic tree. Results Avian influenza virus of Guizhou province H5N1 subtype in duck source consists of a branch, group differences in 0.00-5.00 between avian influenza viruses. Chicken source composed of another branch, group homology, differ- ence is zero. Conclusion Avian influenza virus of Guizhou province has some species-specific in between species. And the avian influenza virus genetic relationship with Hunan province near, between this and two provinces border on geography
