摘  要:目的建立当归地上部分中绿原酸与金丝桃苷的含量测定方法。方法采用高效液相色谱法,色谱柱为GRACE C18(250mm×4.6mm,5μm);流动相为乙腈(A)-0.5%甲酸水溶液(B)梯度洗脱;流速1.0ml/min;检测波长为325nm(绿原酸)和355nm(金丝桃苷)。结果绿原酸和金丝桃苷的线性范围分别为0.29~5.8μg/μL(R2=0.9996),0.1~2.0μg/μL(R2=0.9999);平均加样回收率分别为为97.34%(RSD=4.77%),101.70%(RSD=3.07%)。结论该方法准确可靠、简便可行,可用于当归地上部分药材的质量控制。Objective To establish a HPLC method for the determination of chlorogenic acid and hyperoside in the aerial part of angelica sinensis. Methods The chromatographic separation was achieved on a GRACE C18 ( 250 mm × 4. 6 mm,5 μm) column at a variable wavelength from 325 nm ( Chlorogenic acid) to 355 nm ( hyperoside) using a gradient mobile phase consisted of a mixture of acetonitrile ( A) and 0. 5% formic acid ( B) at a flow rate of 1. 0 ml / min. Results Linearity relationships of chlorogenic acid and hyperoside were 0. 29 ~ 5. 8 μg / μl ( R 2 = 0. 9996 ) ,0. 1 ~ 2. 0 μg / μl ( R 2 = 0. 9999 ) ,respectively; the average recoveries were 97. 34% ( RSD = 4. 77% ) ,101. 70% ( RSD = 3. 07% ) . Conclusion The results showed that the method is accurate,simple,repeatable and it is suitable for the quality control of the aerial parts of Angelica sinensis.
