摘  要:[目的]研究自制多器官保存液(AMU)对兔肾低温保存期间线粒体钙离子内流的阻滞作用.[方法]建立兔离体肾脏单纯低温保存模型,分别采用0~4~C的AMU、离体肾脏保存液(HCA)对兔肾进行灌注和保存,并根据所用保存液的不同分为AMU组和HCA组.分别于低温保存24,48和72h后应用生物化学甲基百里香酚蓝比色法测定肾组织细胞线粒体中钙离子的含量.[结果]随着低温保存时间的延长,AMU组和HCA组肾组织细胞线粒体中钙离子含量均出现增高趋势,两组单纯低温保存兔、肾脏24h时线粒体中的钙离子含量间差异无统计学意义(P〉0.05);保存48,72h时AMU组均明显低于HCA组(P〈0.05).[结论]AMU对兔肾低温保存期间线粒体钙离子内流具有阻滞作用,在保持线粒体钙超载方面优于HCA.OBJECTIVE To study the blockage role of self-designed multiple organ(ATP-MgClurokinase, AMU) preservation solution for mitochondria calcium influx during cold storage of rabbit kidneys METHODS The model of simple cold storage of isolated rabbit kidneys were set up, and the kidneys were flushed and stored with AMU and hypertonic adenine(HCA) preservation solution at 0-4 ℃ , respectively, and after keeping 24 h, 48 h and 72 h, the contents of mitochondrial calcium of renal tissue were measured by using biological and chemical methylthymol blue colorimetric method. RESULTS The contents of mitochondrial calcium were increased both in AMU and HCA groups. The contents of mitochondrial calcium at keeping 24 h had no significant difference between AMU and HCA groups (P〉0.05), but at keeping 48 h and 72 h were significantly lower in AMU group than in HCA group (P 〈 0.05). CONCLUSION AMU preser influx vation solution is superior to HCA preservation solution for blockage role of mitochondria calcium in rabbit kidneys during cold storage
