摘  要:目的观察地佐辛对神经病理性疼痛大鼠行为学及脊髓肿瘤坏死因子-α(TNF-α)和前列腺素E2(PGE2)水平的影响。方法成年雄性大鼠30只随机分为假手术组、模型组和地佐辛组。模型组和地佐辛组大鼠进行坐骨神经慢性压迫性损伤模型制备,假手术组大鼠仅暴露坐骨神经,不进行坐骨神经结扎;地佐辛组大鼠于术后即刻至术后7 d,每日腹腔注射地佐辛5 mg.kg-1,假手术组和模型组大鼠给予等体积的生理盐水。检测术前1 d、术后1、3、5、7、14 d 3组大鼠机械性缩足反射阈值(MWT),并于术后14 d测定大鼠脊髓TNF-α及PGE2水平。结果 3组大鼠术前MWT比较差异无统计学意义(P>0.05),假手术组术后1、3、5、7、14 d大鼠MWT与术前比较差异均无统计学意义(P>0.05),模型组和地佐辛组术后1、3、5、7、14 d大鼠MWT显著低于术前(P<0.05);术后1、3、5、7、14 d,模型组和地佐辛组大鼠MWT均显著低于假手术组(P<0.05),地佐辛组大鼠MWT均显著高于模型组(P<0.05)。模型组和地佐辛组大鼠脊髓中TNF-α和PGE2水平显著高于假手术组(P<0.05),地佐辛组大鼠脊髓中TNF-α和PGE2水平显著低于模型组(P<0.05)。结论地佐辛可抑制坐骨神经慢性压迫性损伤大鼠的神经病理性疼痛,其机制可能与其降低大鼠脊髓中TNF-α和PGE2水平有关。Objective To investigate the influence of dezocine on behavior and the levels of tumor necrosis factor-α(TNF-α) and prostaglandin E2 (PGE2 ) in rats with neuropathic pathologic pain. Methods Thirty adult male rats were ran- domly divided into sham-operation group, model group and dezocine group. The rats in model group and dezocine group were made the model of chronic sciatic nerve oppressive injury; the sciatic nerve of rats in sham-operation group were exposed only, and the sciatic nerve were not ligature& The rats in dezocine group were treated with dezocine 5 mg · kg^-1 by intraperitoneal injection every day from the beginning to seven days after operation;but the rats in sham-operation group and model group were treated with isochoric normal saline. The changes of mechanical withdrawal threshold(MWT) of rats in the three groups were detected at the time of 1 day before operation and 1 day,3,5,7,14 days after operation;14 days after operation, the levels of TNF-α and PGE2 in spinal cord of rats were detected. Results There was no significant difference in MWT among the three groups before treatment(P 〉 0.05). In sham-operation group,the MWTs at 1 day ,3,5,7,14 days after operation had no signifi- cant difference compared with those at pre-operation (P 〉 0.05 ) ;but the MWTs in model group and dezocine group at 1 day ,3,5, 7,14 days after operation were significantly lower than those at preoperation( P 〈 0.05 ). The MWTs in model group and dezocine group were significantly lower than those in sham-operation group( P 〈0.05 ) ,and the MWTs in dezocine group were significantly higher than those in model group at 1 day,3,5,7,14 days after operation( P 〈 0.05 ). The levels of TNF-α and PGE2 in spinal cord of rats in model group and dezocine group were significantly higher than those in sham-operation group( P 〈 0.05 ) ;but the levels of TNF-α and PGE2 in spinal cord of rats in dezocine group were significantly higher than those in model group (P 〈 0. 05)
