摘  要:目的探讨抗肿瘤药物5-氟尿嘧啶(5-Fu)对小鼠外周血及骨髓细胞的影响。方法 80只小鼠随机分为8组,其中4组用于小鼠外周血细胞实验,另4组用于骨髓细胞微核实验;分别为对照组、1/30给药组、1/10给药组和1/3给药组,对照组注射生理盐水0.5mL,其余各组2次腹腔注射不同剂量5-Fu,微核试验观察小鼠外周血和骨髓细胞中微核形成。结果第1次给药后,各组外周血和骨髓细胞中微核率均高于对照组(P<0.01)。第2次给药较第1次给药的微核率明显升高,差异亦有统计学意义(P<0.05)。结论 5-Fu能引起小鼠外周血及骨髓细胞染色体损伤。Objective To investigate the effect of 5-fluorouracil (5-FU) on mouse bone marrow and peripheral blood cells. Methods Eighty mice were randomly divided into 8 groups, and 4 groups used for peripheral blood cells and others for bone marrow. Control group was treated with intraperitoneal injection of normal saline, while 1/30, 1/10 and 1/3 5-FU groups received two intraperitoneal injections of different doses of 5-FU. Micronucleus formation of bone marrow and peripheral blood ceils was evaluated by micronucleus testing. Results Micronucleus formation of bone marrow and peripheral blood cells was increased in 5-FU groups compared with the control group after the first injection (P〈0.01). The second administration induced more micronucleus formation than the first one in 5-FU groups (P〈0.05). Conclusion 5-FU can cause chromosome damage of mouse bone marrow and neriDheral blood cells.
