摘  要:目的探讨血液中四种miRNA在类风湿性关节炎(RA)患者血浆及外周单个核细胞(PBMC)中的表达及意义。方法采用实时荧光定量PCR检测随机选取的43例RA患者和30例健康体检者血浆和PBMC中四种miRNA(miRNA-146a、miRNA-223、miRNA-16和miRNA-155)的表达,分析这些血浆和PBMC特异的miRNA在RA中的作用。结果与对照组相比,RA组患者血浆中miR-146a和miR-155表达下调(P<0.05),miR-16表达增高(P<0.05),而miR-223与健康对照组比较差异无统计学意义(P>0.05);PBMC中四种miRNA表达均上调(P<0.05);相关性分析显示这些miRNA在血浆和PBMC中的表达没有明显相关性。结论 miRNA-146a、miRNA-223、miRNA-16和miRNA-155在RA中起着重要作用且在血浆和PBMC中出现的机制可能不同。Objective To investigate the expression levels of 4 kinds of miRNAs in plasma and peripheral mononuclear cells(PB- MC) of patrients with rheumatoid arthritis(RA) and the clinical significance. Methods Real-time quantitative PCR was adopted to detect the levels of 4 kinds of miRNAs(miRNA-146a,miRNA-223 ,miRNA-16 and miRNA-155) both in plasma and PBMC for the randomly selected 43 cases of RA patients and 30 healthy subjects, and thoses plasma and PBMC specific miRNAs were analysed for their function in RA. Results Compared with control group,in RA group plasma miR-146a and miR-155 were down-regulated(P〈 0.05), miR- 16 expression increased (P〈 0.05), while miR-223 compared with the healthy control group showed no statistically sig- nificant(P〉0.05) ;expression levels of 4 kinds of miRNAs all increased in P MBC (P〈0.05) ;the correlation analysis for the ex- pression levels of miRNAs in plasma and PBMC showed no significant correlation. Conclusion MiRNA-146a,miRNA-223,miRNA- 16 and miRNA-155 play important roles in RA,and the mechanisms for their appearance in plasma and in PBMC may be different.
