摘  要:目的:探讨氯沙坦对心脏保护作用与肝细胞生长因子(HGF)的关系。方法:实验动物为自发性高血压大鼠(SHR)12只,14周龄。分为2组,1组为氯沙坦组(SL),另1组为对照组(SC);以Wistar大鼠为正常对照组(WC),6只,年龄和体质量配比,雌雄配对。SL组每天按30mg/kg灌胃法喂服,对照组采用灌胃法喂服等量蒸馏水,疗程6周。实验结束后分别测量血压和左室质量,采用M-30-apoptosense ELISA法测心肌细胞凋亡数量及血浆和组织HGF浓度。结果:治疗6周后,与对照组(SC)相比,SL组血压下降,左室质量指数降低,心肌细胞凋亡数量减少,血浆HGF降低,伴有组织HGF浓度升高。结论:氯沙坦能够促进组织HGF分泌,逆转心室肥厚,减少心肌细胞凋亡。Objective To study the relationship of hepatocyte growth factor(HGF) and cardioprotectionof Losartan during left ventricular remodeling in SHR. Methods 12 SHRs aged 14 weeks were randomly divided into two groups and treated for 6 weeks as follows Losartan 30mg/kg/day, n = 6; distilled water, n = 6; compared with 6 WKYs aged 14 weeks. Systolic blood pressure was measured by the tail-cuff method each week. After treatment, the rats were killed. Blood samples were collected by decapitation without anesthesia in prechilled tubes and were stored at-70℃ before measurement. The cardiac tissue were promptly removed from the apex and frozen in liquid nitrogen and then stored at-70℃ until use. The HGF of plasma and myocardium were determined by ELISA. Cardiomyocyte apoptosis was identified by M-30-apoptosense TMELISA. Results (1)Sysdtolic blood pressure, left ventricular weight index and cardiomyocyte apoptosis all significantly decreased in SHR-Los artsn compared with SHR-control group. Compared with SC group, plasma HGF also strongly decreased at SHR-Losartan group, but HGF in the cardiac tissue increased obviously. Conclusion Losartan can effectively reverse left ventricular hypertrophy of SHR and decrease the cardiomyocyte apoptosis via inducing tissue HGF production.
