银杏酮酯对抑郁模型大鼠行为学及血清皮质酮的影响

摘　　要：目的观察银杏酮酯(GBE50)对抑郁模型大鼠行为学的影响,并检测各组大鼠血清皮质酮浓度,探讨GBE50抗抑郁的效果。方法采用国内外实验室普遍应用的慢性不可预见性应激抑郁模型。将60只雄性SD大鼠随机分为6组,包括正常对照组,模型组,西药(氟西汀)组,中药(GBE50)高、中、低剂量组。经21天应激结合同时给药,对各实验组进行强迫游泳实验测试并检测各组血清皮质酮含量。结果与正常对照组比较,模型组大鼠挣扎时间明显缩短(P<0.01),不动时间则显著延长(P<0.01);与模型组比较,中药高、中剂量组可显著延长大鼠挣扎时间(P<0.01),西药组和中药高、中剂量组可明显缩短大鼠不动时间(P<0.01)。与正常对照组比较,模型组大鼠血清皮质酮浓度明显增加(P<0.01);与模型组比较,氟西汀组及中药高、中剂量组大鼠的血清皮质酮浓度均显著下降(P<0.01)。结论 GBE50可显著缓解模型大鼠抑郁样行为,提示GBE50具有一定治疗抑郁症作用。Objective To investigate the anti-depression effect of Ginkgo biloba extract 50 （GBES0） by observing its effect on behavioristics and detecting serum corticosterone level in rats. Methods The models were established by chronic unpredictable stress. Sixty male SD rats were randomly divided into six groups： control group, model group, Fluoxetine group and GBES0 groups （large, medium and small dose）. After oral administration for 21 days, the serum cortieosterone level was detected by forced swim test. Results Compared with normal control group, struggling time in model group was significantly decreased （ P 〈 0.01 ） , while immobility time in model group was increased （ P 〈 0.01 ） ; compared with model group, struggling time was obviously increased in GBES0 large and medium dose groups （P 〈0.01 ）, and immobility time was obviously shortened in Fluoxetine group and GBE50 large and medium dose groups （P 〈0.01 ）. The serum corticosterone level in model group was significantly higher than that of the control group （P 〈 0.01 ） ; while the levels in Fluoextine group, GBE50 large and medium groups were significantly reduced than that of the model group （P 〈 0.01 ）. Conclusion GBE50 can relieve the depressed-like behavior in model rats. The results suggest that GBE50 has positive effects on depression.
