摘 要:目的观察双黄糖脂宁片去蒽醌提取后对自发性2型糖尿病(GK)大鼠糖脂代谢紊乱的影响。方法选用10只健康雄性Wistar大鼠作为正常组,将36只GK大鼠根据空腹血糖(FPG)并参考糖负荷2小时血糖(2hPG)分为模型组、中药低剂量组、中药高剂量组和二甲双胍组,每组9只,予饮用水或药物灌胃4周;4周时测FPG、2hPG、随机血糖(RPG),检测血脂4项[总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)和高密度脂蛋白胆固醇(HDL-C)]、糖化血红蛋白(HbAlc)和肝脏三酰甘油(L-TG),检测血清胰岛素(Ins)并计算胰岛素敏感指数(ISI)。结果与正常组比较,模型组大鼠FPG、2hPG、RPG、HbAlc、血清TC、LDL-C、L-TG均显著升高,Ins、ISI显著降低(P<0.01)。与模型组比较,各用药组均能显著提升ISI水平、降低HbAlc水平(P<0.05);中药各剂量组RPG、L-TG水平均显著降低(P<0.01);中药低剂量组TG水平显著降低(P<0.05);中药高剂量组Ins、TC水平显著降低(P<0.01);二甲双胍组RPG、TG、L-TG、ISI亦显著降低(P<0.05)。结论去蒽醌双黄糖脂宁片能显著改善GK大鼠糖脂代谢紊乱,具有良好的开发前景。Objective To observe the clinical efficacy of "Shuanghuang Tangzhining Tablet" without Anthraquinone on glucose and lipid metabolism disorder in GK rats. Methods Ten healthy male Wistar rats were selected as normal group. Thirty-six Goto-Kakizaki rats with type 2 diabetic were divided into model group, Chinese medicine groups ( small and large dose) and DMBG group according to FBG and 2 hPG, with 9 rats in each group. The rats were administrated intragastrically with water and medicine for 4 weeks. After 4 weeks the FPG, 2 hPG, RPG, TC, TG, LDL-C, HDL-C, HbAlc, L-TG and Ins were detected and the ISI was calculated by "Liguangwei Method". Results Compared with normal group, the FPG, 2hPG, RPG, HbAlc and TC, LDL-C, L-TG in the serum of the model group were increased significantly, while the Ins and ISI were decreased markedly (P 〈 0.01 ). Compared with model group, the ISI in all the medicine groups were increased markedly and HbAlc level was decreased ( P 〈 0.05 ) ; the RPG level and L-TG level in Chinese medicine groups were decreased significantly (P 〈0.01 ) ; the TG level in small dose Chinese medicine group was decreased markedly (P 〈 0.05 ) ; the Ins level and TC level in large dose Chinese medicine group were 'decreased markedly ( P 〈 0.01 ) ; the levels of RPG, TG, L-TG and ISI in DMBG group were decreased significantly ( P 〈 0.05 ). Conclusion "Shuanghuang Tangzhining Tablet" without Anthraquinone can improve the glucose and lipid metabolism disorder in GK rats significantly. It has a good development prospects.
