摘  要:目的应用HPLC-HR-MSn法快速鉴定苦碟子注射液中的酚酸类成分。方法采用Thermo Hypersil BDS C18(4.6 mm×250 mm,5μm)色谱柱,以0.1%甲酸-乙腈溶液为流动相,梯度洗脱;应用ESI-LTQ-Orbitrap质谱负离子模式检测苦碟子注射液中的酚酸类化合物。结果苦碟子注射液中酚酸类成分HPLC分离良好,根据多级质谱和精确分子量数据,从中共鉴定了25个酚酸类成分,包括绿原酸类化合物15个,酒石酸类化合物6个以及小分子酚酸4个。结论建立了酚酸类成分的HPLC-HR-MSn检测方法,为进一步阐明苦碟子注射液药效物质基础提供科学依据。Objective To identify the phenolic acids in Kudiezi injection by HPLC-HR-MSn quickly.Methods Thermo Hypersil BDS C18 column(4.6 mm×250 mm,5 μm) was used with 0.1% formic acid-acetonitrile solution as the mobile phase in gradient elution.The phenolic acids were detected by ESI-LTQ-Orbitrap equipped with an ESI ion source in a negative mode.Results Based on the MS/MS fragmentation behaviors and high-resolution mass information,25 phenolic acids were identified,15 of which were chlorogenic acids and its derivatives,6 were tartaric acids,and the other 4 were micromolecules of phenolic acids.Conclusion The established method of detecting similar compounds by mass spectrometry is useful for further study of pharmacodynamic substance from Kudiezi injection.
