尿激酶溶解鼠颈总动脉血栓的动态观察

摘　　要：目的 :探讨尿激酶溶栓治疗的时间窗。方法 :分离 SD鼠颈总动脉 ,用 50 % Fe Cl3制作颈总动脉血栓模型 ,分别于动脉血栓形成后 0 .5、1 .0、2 .0、3.0、6.0、8.0小时静脉注射尿激酶 30 ,0 0 0 U/kg,观察不同用药时间 SD鼠颈总动脉血栓的溶解过程 ,记录血栓开始溶解时间及血栓完全溶解时间。结果 :动脉血栓形成后 0 .5、1 .0、2 .0、3.0、6.0小时静脉注射尿激酶 ,血栓开始溶解时间分别是病程约 50分钟、2小时、3.5小时、4 .5小时、8.0小时 ;血栓完全溶解时间分别是病程约 1 .5小时、3小时、4 .5小时、6小时、 1 0小时。而动脉血栓形成后 8小时静脉注射尿激酶 ,观察 2 4小时未见血栓溶解。结论 :急性脑梗死最佳溶栓治疗时间窗是病程 3小时以内。Objective: To investigate the time window of urokinase thrombolysis .Methods: The carotid thrombosis model was made in SD rats by using 50% FeCl 3 Urokinase (30000U/kg) was injected intravenously at 0.5th at 0.5th、1.0th、2.0th、3.0th、6.0th、8.0th hour after thrombosis respectively .Both beginning time and complete time of thrombolysis was recorded and the course of thrombolysis was observed at different injection time of urokinase after thrombosis.Results: The beginning time of thrombolysis at 0.5th、1.0th、2.0th、3.0th、6.0th、8.0th hour by injecting urokinase after thrombosis were 50 mim、2、3.5、4.5 and 8 hours respectively. The time of complete thrombolysis were 1.5、3、4.5、6 and 10 hours respectively. No thrombolysis was found during 24 hours after urokinase injection at 8th hour of thrombosis .Conclusion: The optimal time window of urokinase thrombolysis in the acute ischemic stroke is less than 3 hours after thrombosis.
