摘 要:目的探讨甲醛和β-丙内酯灭活轮状病毒的效果。方法采用甲醛和β-丙内酯灭活轮状病毒,通过微量滴定法检测灭活病毒滴度变化,盲传3代验证病毒灭活效果,电镜检测灭活轮状病毒的结构,ELISA评价灭活轮状病毒诱发小鼠血清抗体水平等。结果甲醛灭活72 h和β-丙内酯灭活24 h,轮状病毒被完全灭活;灭活的病毒结构保持完整,免疫小鼠可诱发血清轮状病毒特异性抗体,且与灭活前无显著差异。结论甲醛和β-丙内酯均可有效地灭活轮状病毒。Objective To investigate the effect of inactivation with formaldehyde(F) and β-propiolactone(BPL) on rotavirus.Methods The rotavirus was inactivated by F and BPL,the change of infectious titers of inactivated rotavirus was measured by microtitrimetry.The effect of inactivation was verified by blind passage three times.The structure of inactivated rotavirus was detected by electron microscope.The level of antibody induced by inactivated rotavirus in mice was evaluated by ELISA.Results Rotavirus was completely inactivated after treatment with F for 72 h or BPL for 24 h.The structure of rotavirus inactivated by F and BPL kept integrity.The serum rotavirus specific antibody in mice was induced after immunization with rotavirus inactivated by F and BPL,and it was no significant difference in the level of antibody between before and after inactivation.Conclusion Rotavirus can be effectively inactivated by both F and BPL.
