摘 要:目的探讨β-七叶皂甙钠对肠缺血-再灌注损伤早期大鼠存活率的影响。方法随机分为:①假手术组;②模型组;③治疗组。制作肠缺血-再灌注损伤模型。Chiu’s法观察肠粘膜损伤情况,电镜观察肠粘膜超微结构变化;测定粘膜血管通透性、组织髓过氧化物酶(MPO)活性与降钙素基因相关肽(CGRP)含量;观察大鼠存活率。结果小肠缺血再灌注后,模型组肠粘膜屏障严重受损、β-七叶皂甙钠能减轻其损伤。β-七叶皂甙钠能降低Chiu’s评分、EB含量、MPO活性与CGRP含量,并提高大鼠的存活率。结论预先给予β-七叶皂甙钠能提高缺血-再灌注损伤大鼠存活率,可能与其对肠粘膜屏障形态与功能保护有关。Objective The aim of study was to observe the influence of β-aescinate sodium on the survival rates after intestinal ischemia-reperfusion injury in rats.Methods Rats were randomly divided into three groups:the sham group,the model group,the sodium aescinate group.Intestinal ischemia reperfusion injury was established.The histopathological changes of intestinal mucosa were examined by HE staining,The ultrastructure of gut barrier was observed with electron microscope.Intestinal vascular permeability,the activity of myeloperoxidase and the content of CGRP were measured.the survival rates were observed.Results The gut barrier of the rats was seriously damaged on transmission electron microscope in group M and was attenuated by sodium aescinate treatment.The restitution of gut barrier function was obviously slower than that of the morphology and there was no direct correlation between them.Sodium aescinate could attenuate the up-regulation of the Chiu's score and subtract the vascular permeability,the activity of MPO and the content of CGRP(P0.05),and increase the survival rates.Conclusion Pretreated of sodium aescinate prior to reperfusion may increase the survival rates against ischemia-reperfusion damage,which is associated with its protection on the morphology and function of intestinal mucosal barrier.
