摘 要:目的为获得临床对猪大肠杆菌病较好的预防方法,研究猪产毒素大肠杆菌K88黏附素FaeG基因的重组乳酸菌对小鼠免疫性能的影响。方法将重组乳酸菌经口服(灌胃)免疫BALB/C小鼠,加强免疫后7 d处理小鼠,采集血液、脾脏、粪便,处理后采用试剂盒及间接ELISA方法,检查小鼠免疫性能变化情况。结果重组乳酸菌经口服免疫小鼠后,提高了小鼠生长初期的脾脏指数及细胞因子(IL-2)的含量,加强免疫后粪便中的sIgA与对照组相比,差异极显著(P<0.01),血清中猪ETEC(K88)的特异性抗体水平高于对照组,差异性达到了统计学上的显著水平(P<0.05)。结论含有猪ETEC K88黏附素FaeG基因的重组乳酸菌,可以诱导小鼠产生良好的针对猪产毒素大肠杆菌的全身体液和黏膜免疫应答。Objective Enterotoxigenic Escherichia coli (ETEC) are major pathogens causing diarrhea in newborn piglets and calves. In an attempt to get a better approach to clinically prevent colibacillosis of pigs.Method As an oral vaccine, the recombinant Lactobaeillus was administered to BALB/C mice by gastric perfusiun. After immunizing the mice for seven days, samples of blood, spleen and excrement were collected to detect immune Performances of the exogenous gene expressed in immunized mice. Result As measured by Kits and indirect ELISA, mice receiving gastric perfusion exhibited a better spleen index and higher IL-2 level when compared with the control group. Furthermore, the immunized mice revealed a statistically significant increase in specific antibody for ETEC K88 in blood serum (p〈O.05) and sIgA in faeces (p〈0.01). Conclusion These resuhs indicate the crucial functions of the genetically modified Lactobacillus containing ETEC K88 fimbrial adhesion FaeG gene in inducing evident humoral and mucosal immune Performances to ETEC in mice.
