摘　　要：目的血型鉴定正确是临床输血安全的前提。文中对一种新研制的ABO反定型免疫胶体金试剂进行评估。方法应用胶体金标记A、B和O型红细胞膜抗原,制备ABO反定型免疫胶体金试剂;利用试管法和微量板法对其特异性、亲和力、稳定性、灵敏度及准确率进行评估。结果本试剂的特异性和亲和力均符合常规使用要求,且稳定性高,可在2～8℃储存1年,其灵敏度与传统反定型红细胞相当。对809份临床样本进行血型反定型检测,准确率达99.5%。结论 ABO反定型免疫胶体金试剂可以用于常规血型鉴定。Objective Correct grouping is a prerequisite for safe blood transfusion. The aim of this study was to evaluate the properties of a newly developed immunocolloidal gold reagent for ABO reverse typing. Methods The immunocolloidal gold re- agent for ABO reverse typing was prepared through colloidal gold labeling A, B and O red cell membrane antigens. Then the speci- ficity, affinity, stability, sensitivity, and accuracy of this immunocolloidal gold reagent were assessed using the tube test and micro- plate test. Results Both the specificity and affinity of this immunocolloidal gold reagent met the requirements of routine use. Fur- thermore, it exhibited a high stability and could be stored at 2 to 8℃ for 1 year, with a sensitivity comparable to that of traditional reagent red cells in reverse ABO typing. The accuracy rate of the immunocolloidal gold reagent was 99.5% in 809 clinical samples. Conclusion The immunocolloidal gold reagent for ABO reverse typing can he used in routine blood typing.
