摘 要:目的:建立适用于MRI瘤周浸润研究的鼠脑胶质瘤模型。方法:通过立体定向技术,将c6细胞接种至A、B两组大鼠右侧尾状核,对照组在相同部位注射全培养基。A、B两组荷瘤大鼠分别于接种后1~2和3~4周利用3.0T磁共振扫描仪行常规MRI、T1wI增强扫描。病理观察肿瘤级别及肿瘤浸润程度,分析A、B两组肿瘤细胞浸润程度差别。结果:49只存活的细胞接种大鼠,在随后的MRI或病理检查中证实有肿瘤形成,对照组大鼠0=5)未见肿瘤形成。磁共振成像显示B组瘤体较A组体积大,信号欠均匀,增强后呈明显均匀强化或环状强化。A、B组荷瘤大鼠瘤周浸润程度分别平均为(0.083士0.005)和(0.315±0.012),两组结果经两样本,检验显示差别有统计学意义(t=11.8,P〈0.05)。结论:肿瘤细胞接种后3~4周是肿瘤细胞浸润最明显的阶段,可作为MRI脑胶质瘤浸润研究的理想模型。Purpose: To establish the stable and reliable rat brain glioma model for MRI study of glioma peritumoral invasion. Methods: C6 glioma cells and whole culture medium were stereotactically injected into the right caudate nucleus of the experimental group (group A, B) and control group, respectively. About 1-2 weeks and 3-4 weeks after implantation, all survival rats were examined by conventional MRI, enhanced TIWI scans using a GE 3.0T MRI scanner, respectively. Tumor grade and degree of tumor infiltration were observed under the microscope. And difference of tumor infiltration degree was analyzed between group A and B. Results: Forty-nine survival rats were confirmed with the tumor formation by the subsequent MRI scans or pathological examination. And rats in control group (n=5) were with no tumor formation. Magnetic resonance imaging showed that tumor volumes of rat brain glioma were larger in group B than that in group A, and the tumors were characterized by inhomogeneous signal and homogeneous enhancement or ring enhancement. Degree of tumor infiltration in group A and B was (0.083±0.005) and (0.315±0.012), respectively. The difference was with statistical significant between group A and B (t=11.8, P〈0.05). Conclusion: The rat brain glioma model is in the most significant stage of tumor cell infiltration on about 3 to 4 weeks after inoculation, and it can be used as the ideal model for MRI study of brain glioma infiltration.
