摘 要:目的:比较不同脱细胞方法制备的大鼠坐骨神经细胞外基质材料(acellular extracellular matrix,AECM)的组分,为解决临床周围神经损伤修复移植体的来源提供实验依据。方法:分别采用优化的化学萃取脱细胞方法和低渗-酶消化法制备大鼠坐骨神经AECM,通过HE染色、变色酸2R-亮绿染色、免疫组织化学染色和SDS-PAGE对AECM的结构成分进行检测和比较。结果:不同方法制备的大鼠坐骨神经AECM在基底膜管完整性、细胞外基质(extracellular matrix,ECM)活性、去细胞程度和脱髓鞘程度方面无显著性差异。结论:应用优化的化学萃取脱细胞方法和低渗-酶消化法制备的AECM,均具有良好的仿生性,是修复周围神经缺损的适宜移植物。Objective:By comparing the composition of acellular extracellular matrix (AECM) obtained via different decellulariza- tion protocols, we aim at supplying experimental evidence for the solution of the source of grafts in clinical peripheral nerve repairs. Methods :The transplants prepared through optimized chemical processing and with hypotonic - enzymatic method were obtained from rats. The construction and composition of AECM were evaluated by performing HE dyeing, chromotropic acid 2R - brilliant green stai- ning, immunohistochemistry and SDS - PAGE. Results:There was no significant difference in the integrity of vessel of basilar mem- brane, the activity of extracellular matrix ( ECM ) and the extent of removal of cells and myelin between different decellularization proto- cols. Conclusion: AECM obtained via optimized chemical processing and hypotonic - enzymatic method in rats appeared to be bionic, thus both scaffolds could be used as suitable transplants for effective repair of peripheral nerves.
