摘  要:目的:研究抵当汤对高脂饮食胰岛素抵抗大鼠的瘦素(LP)、脂联素(APN)及其受体mRNA表达的影响。方法:大鼠35只随机分为5组,空白对照组每日给予普通饲料和蒸馏水(10ml/kg)灌胃;模型组每日给予高脂饲料和蒸馏水(10ml/kg)灌胃;抵当汤低剂量组每日给予高脂饲料,低剂量抵当汤药液灌胃(1g/kg);抵当汤高剂量组每日给予高脂饲料,高剂量抵当汤药液灌胃(2g/kg);血脂康组每日给予高脂饲料,血脂康灌胃(0.2g/kg)。连续喂养8周,留取血清、血浆标本,进行生化指标及血清脂联素及血清瘦素的检测;迅速切取部分肝脏组织,进行脂联素及瘦素受体表达的检测。结果:模型组与空白组相比,大鼠脂联素均明显降低(P<0.01),瘦素增多(P<0.05);肝脏脂联素受体AR2mRNA)表达降低(P<0.05);瘦素受体OB-RmRNA表达明显降低(P<0.01);抵当汤低、高剂量组与模型组相比,大鼠APN增多(P<0.01),抵当汤低、高剂量组LP降低(P<0.05);抵当汤高剂量组大鼠AR2mRNA表达显著增多(P<0.01),抵当汤低剂量组AR2mRNA表达增多(P<0.05);抵当汤高剂量组大鼠OB-R mRNA表达明显增多(P<0.01)。结论:抵当汤可提高胰岛素抵抗大鼠胰岛素敏感性,防治胰岛素抵抗;抵当汤可升高胰岛素抵抗大鼠血清脂联素水平,降低血清瘦素水平;抵当汤可上调胰岛素抵抗大鼠肝脏脂联素受体AR2mRNA、瘦素受体OB-RmRNA的表达。Objective: To study the effects of Didangtang on the expression of adiponectin and leptin and their receptors mRNA in rat model with insulin-resistant induced by high fat diet.Methods: 35 health and clean level Wistar male rats were chosen and randomly divided into 5 groups: control group,model group,low-dose Didangtang group,high-dose Didangtang group and Xuezhikang group with 7 rats each.The rats of control group were fed with normal diet and distilled water(10ml / kg) daily,model group was fed high fat diet and distilled water(10ml/kg) daily,low-dose Didangtang group was fed high fat diet and Didingtang(1g/kg) daily,high-dose Didangtang group was fed high fat diet and Didingtang(2g / kg) daily,Xuezhikang group was fed high fat diet and Xuezhikang(0.2g / kg) daily.After 8 weeks of feeding,take blood by decapitate.Blood was taken to measure biochemical markers and serum adiponectin and leptin.The liver tissue was taken promptly to measure AR2mRNA,OB-R mRNA.Results: Didangtang can reduce the IR rats serum leptin levels,raise serum adiponectin levels and increase the expression of adiponectin receptor and leptin receptor in the liver tissue of IR rats.Conclusion: Didangtang can improve insulin sensitivity.Improve insulin resistance.Didangtang is an ideal medicine in preventing and treating dyslipidemia.
