五苓散对EG-NH_4 Cl诱导大鼠肾结石的影响

摘　　要：目的：研究五苓散防治雄性大鼠草酸钙肾结石形成的作用,为其临床治疗结石症提供实验依据。方法：大鼠随机分为：正常组（Normal）,模型组（Model）,枸橼酸钾组（PC,2.5g/kg）,五苓散低剂量（LW,4.8g/kg）和高剂量组（HW,9.6g/kg）,共5组。采用1%乙二醇和0.5%氯化铵混于饮用水中制备大鼠草酸钙肾结石模型,同时各组给予相应药物,每天测量大鼠体重和饮水量,连续给药4周后,检测大鼠24h尿量及尿液结石情况,测定尿pH、Ca2＋、Mg2＋、Ox含量和大鼠肾功能,血清Ca2＋、Mg2＋含量;HE染色及偏光显微镜检测各组大鼠肾组织病理学改变和草酸钙结晶沉积情况。结果：五苓散在剂量4.8~9.6g/kg剂量范围内,均可降低血钙、尿草酸等促肾结石形成物质的含量,增加尿石形成抑制物镁的含量,促进肾结石溶解,明显抑制尿中和肾组织中草酸钙晶体的形成及减轻肾功能损伤程度。结论：五苓散可明显抑制草酸钙晶体的形成,保护肾脏。Objective： To provide experimental basis for clinical application of Gorei-San through studying on preventing and treating renal stone in rats.Methods： Forty male Sprague-Dawley rats,weighing 180g ~ 220g,were randomly divided into five group,the normal goups（Normal）,the model group（Model）,the potassium citrate group（PC,2.5g / kg）,Gorei-San low dose group（LW,4.8g / kg）,Gorei-San high dose group（LW,9.6g / kg）.Renal stone model were produced by free access to 1% EG plus 0.5% NH 4 Cl in drinking water.PC（2.5g / kg,ig） and Gorei-San（4.8g / kg,9.6g / kg,ig） were administered daily for 4 weeks.The water consumption and weight of rats were recorded every day.The level of Cr in serum was tested by autobiochemical analyser,Ca2 ＋and Mg2 ＋in serum and the content of Ca2 ＋,Mg2 ＋,Ox in urine were measured by ion chromatography.The pathomorphological changes to the renal tissue were also carried out to probe the effects of GoreiSan.Results： Compared to the model group,the dose range from 4.8g / kg to 9.6g / kg of Gorei-San treatment decreased the concentration of Ca and Ox,increased the content of Mg2 ＋,dissolved kidney stones and lessen formation of calcium oxalate crystal and injury of renal cell.Conclusion： Gorei-San could inhibit calcium oxalate stone formation in experimental rats,showing a great protection effect on kidney.
