桂枝茯苓胶囊对大鼠慢性盆腔炎的影响

摘　　要：目的:观察桂枝茯苓胶囊对慢性盆腔炎大鼠炎症细胞因子的影响,探讨桂枝茯苓胶囊治疗慢性盆腔炎的作用机制。方法 :采用15%苯酚胶浆复制大鼠慢性盆腔炎模型,造模15 d后,随机分为假手术组,模型组,桂枝茯苓胶囊低、中、高剂量组(0.5,1.0,2.0 g/kg)和妇科千金胶囊组(1.6 g/kg)。各治疗组灌胃给药30 d,观察大鼠子宫内膜的病理变化,测定大鼠血清肿瘤坏死因子-α(TNF-α)、白介素-2(IL-2)含量。免疫组织化学法检测各组大鼠子宫细胞间粘附分子-1(ICAM-l)表达。结果:桂枝茯苓胶囊能明显改善慢性盆腔炎大鼠子宫内膜病变程度,与假手术组大鼠相比,模型组大鼠血清IL-2明显降低,TNF-α明显升高,子宫组织中ICAM-l表达显著升高。经桂枝茯苓胶囊治疗后,大鼠血清IL-2较模型组明显增高,TNF-α明显降低,ICAM-l表达显著显著降低。结论:桂枝茯苓胶囊对慢性盆腔炎症具有治疗作用,作用机制可能与调节炎症细胞因子有关。Objective： To probe the mechanism of Guizhi Fuling Capsule for treatment of chronic pelvic inflammation through observing inflammatory cytokine in serum.Methods： The animal model of chronic pelvic inflammation was replicated by injection of 15 % phenol mucilage.After 15 days,The rats were divided into six groups randomly,normal group,model group,lower dosage of Guizhi Fuling capsule（0.5 g / kg）,middle dosage of Guizhi Fuling capsule（1.0 g / kg）,large dosage of Guizhi Fuling capsule（2.0 g / kg）,and Qianjing capsule（1.6 g / kg）.After the rats were treated 30 days with corresponding medicine by intragastric administration,the pathological of endometrium was observed and the concentrations of tumor necrosis factor（TNF-α）,interleukin-2（IL-2） were examined in serum.The expression of the ICAM-l in chronic pelvic inflammatory disease rats were measured by immunohis-tochemistry.Results： Guizhi Fuling capsule could alleviate obviously the pathological damage of endometrium.In comparison with sham operated group,serum IL-2 in model group was significantly decreased and TNF-α was significantly elevated and the expression of the ICAM-l in chronic pelvic inflammatory disease rats were significantly elevated.After Guizhi Fuling capsule treatment,serum IL-2 was obviously elevated compared with the model group and TNF-α was obviously decreased and ICAM-l was obviously decreased.Conclusion ： Guizhi Fuling capsule can alleviate the inflammation of pelvic by regulating the expression of inflammatory cytokines in serum.
